【摘要】：目的觀察雷帕霉素對(duì)缺血再灌注損傷(RIRI)大鼠視網(wǎng)膜組織結(jié)構(gòu)、內(nèi)層厚度的影響,并探討其機(jī)制。方法將65只健康成年SD大鼠隨機(jī)分為對(duì)照組5只、模型組30只和觀察組30只。模型組和觀察組采用前房灌注法制備RIRI大鼠模型,觀察組大鼠造模前2 h腹腔注射雷帕霉素(2 mg/kg),模型組大鼠腹腔注射等量的生理鹽水。對(duì)照組不作任何處理。造模后0、2、6、12、24、48 h模型組、治療組分別處死5只大鼠,對(duì)照組5只大鼠實(shí)驗(yàn)結(jié)束后處死,摘除眼球制成石蠟切片。HE染色光鏡下觀察各組大鼠視網(wǎng)膜組織形態(tài),測(cè)量視網(wǎng)膜內(nèi)層厚度;用免疫組化法檢測(cè)各組大鼠視網(wǎng)膜組織中的Caspase-7、Calpain-1表達(dá)的平均光密度(AOD)。結(jié)果模型組大鼠造模后早期視網(wǎng)膜水腫增厚,晚期各層細(xì)胞排列疏松,視網(wǎng)膜神經(jīng)節(jié)細(xì)胞數(shù)目減少,發(fā)生空泡樣變性。觀察組大鼠視網(wǎng)膜損傷程度和視網(wǎng)膜神經(jīng)節(jié)細(xì)胞空泡變性程度均低于模型組相同時(shí)點(diǎn)。造模后0、2 h觀察組視網(wǎng)膜內(nèi)層厚度小于模型組(P均0.05),造模后6、12、24、48 h觀察組大鼠視網(wǎng)膜內(nèi)層厚度大于模型組(P均0.05)。造模后各時(shí)點(diǎn)觀察組視網(wǎng)膜組織Caspase-7、Calpain-1的AOD均低于模型組相同時(shí)點(diǎn)(P均0.05),但均高于對(duì)照組(P均0.05)。結(jié)論雷帕霉素可以減輕RIRI大鼠視網(wǎng)膜損傷程度,其機(jī)制可能與其降低視網(wǎng)膜組織Caspase-7、Calpain-1表達(dá)有關(guān)。
[Abstract]:Objective to observe the effects of rapamycin on retinal tissue structure and inner layer thickness in rats with ischemia-reperfusion injury (RIRI) and to explore its mechanism. Methods 65 healthy adult SD rats were randomly divided into control group (n = 5), model group (n = 30) and observation group (n = 30). The RIRI model was established in the model group and the observation group by anterior chamber perfusion. The rats in the observation group were intraperitoneally injected with rapamycin 2 mg / kg of rapamycin 2 mg 路kg ~ (-1) before the model was made, and the model group was injected with the same amount of normal saline. No treatment was given in the control group. 5 rats were killed in the treatment group and 5 rats in the control group were killed after the experiment. The eyeballs were removed and made into paraffin section. The morphology of retinal tissue was observed under light microscope and the thickness of the inner layer of retina was measured under light microscope. The average optical density of Caspase-7 calpain-1 expression in the retina of rats in each group was detected by immunohistochemical method. Results in the model group, the retinal edema thickened in the early stage, the cells in the late stages were loose, the number of retinal ganglion cells decreased, and vacuolar degeneration occurred. The degree of retinal injury and degeneration of retinal ganglion cells in the observation group were lower than those in the model group at the same time point. The thickness of the inner retinal layer in the observation group was lower than that in the model group (P < 0.05) 2 hours after the model making, and the thickness of the inner retinal layer in the observation group was greater than that in the model group (P < 0.05). The AOD of Caspase-7 calpain-1 in the retina of the observation group was lower than that of the model group at the same time points (P < 0.05), but higher than that of the control group (P = 0.05). Conclusion rapamycin can attenuate the degree of retinal injury in RIRI rats, and its mechanism may be related to the decrease of Caspase-7 calpain-1 expression in retinal tissues.
