異甘草素對人鼻咽癌CNE2細(xì)胞裸鼠移植瘤模型的放療增敏作用

【摘要】:目的觀察異甘草素對鼻咽癌裸鼠移植瘤放療的增敏效應(yīng),并探討其與乏氧誘導(dǎo)因子-1α(HIF-1α)、血管內(nèi)皮生長因子(VEGF)表達(dá)水平之間的關(guān)系。方法取低分化鼻咽癌CNE2細(xì)胞接種于雌性裸鼠皮下(共接種20只),待腫瘤結(jié)節(jié)4~6 mm時,將裸鼠隨機(jī)分為4組,分別為對照組、異甘草素組(50 mg·kg~(-1)·周~(-1))、放療組、異甘草素(50 mg·kg~(-1)·周~(-1))聯(lián)合放療組。依照各組不同治療措施處理,定期測量瘤體體積,20 d后處死裸鼠,剝離瘤體稱重。分別計(jì)算4組抑瘤率。采用熒光定量PCR檢測瘤體組織中HIF-1α及VEGF mRNA的表達(dá)水平。結(jié)果異甘草素聯(lián)合放療組移植瘤的體積和重量顯著下降,與對照組、單純異甘草素及單純放療組比較差異均有統(tǒng)計(jì)學(xué)意義(P0.05);異甘草素聯(lián)合放療組HIF-1α、VEGF mRNA表達(dá)水平明顯下降,與對照組及放療組相比,差異有統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論異甘草素能增強(qiáng)鼻咽癌裸鼠移植瘤對放療的敏感性,其作用機(jī)制可能與抑制HIF-1α、VEGF表達(dá)水平,從而抑制腫瘤血管生成作用有關(guān)。
[Abstract]:Objective to observe the sensitizing effect of isoglycyrrhizin on radiotherapy of transplanted nasopharyngeal carcinoma (NPC) in nude mice, and to investigate its relationship with the expression of hypoxia inducible factor 1 偽 (HIF-1 偽) and vascular endothelial growth factor (VEGF). Methods CNE2 cells of poorly differentiated nasopharyngeal carcinoma (NPC) were inoculated subcutaneously into female nude mice (20 female nude mice). When tumor nodules were 4mm, nude mice were randomly divided into 4 groups: control group, isoglycyrrhizin group (50 mg 路kg ~ (-1) and radiotherapy group. Isoglycyrrrhizin 50 mg / kg / kg / 1) combined with radiotherapy group. According to the different treatment measures, the tumor volume was measured regularly for 20 days, then the nude mice were killed and the tumor was weighed. Tumor inhibition rates were calculated in 4 groups. The expression of HIF-1 偽 and VEGF mRNA in tumor tissue was detected by fluorescence quantitative PCR. Results the volume and weight of xenografts were significantly decreased in the combined radiotherapeutic group. There were significant differences in the volume and weight of xenografts compared with those in the control group, the simple glycyrrhizin group and the radiotherapy alone group (P 0.05), and the expression of HIF-1 偽 mRNA in the radiation group was significantly lower than that in the control group. Compared with the control group and radiotherapy group, the difference was statistically significant (P 0.05). Conclusion Isoglycyrrhizin can enhance the radiosensitivity of nasopharyngeal carcinoma xenografts to radiotherapy in nude mice, and its mechanism may be related to the inhibition of the expression of HIF-1 偽 -VEGF, and thus the inhibition of tumor angiogenesis.
