眼瞼鱗癌和基底細(xì)胞癌中激活蛋白AP-1、程序性細(xì)胞死亡因子PDCD4蛋白與p73蛋白表達(dá)的研究

【摘要】:目的觀察AP-1(激活蛋白-1)、PDCD4(程序性細(xì)胞死亡因子-4)與p73蛋白在眼瞼鱗癌不同病理分期和基底細(xì)胞癌的表達(dá)及臨床意義,并探討三者與眼瞼基底細(xì)胞癌和鱗癌發(fā)生發(fā)展的關(guān)系。 方法實(shí)驗(yàn)研究:用免疫組化染色方法檢測68例眼瞼鱗癌和40例基底細(xì)胞癌,并以20例正常眼瞼組織作對照,檢測三類組織中AP-1、 PDCD4與p73蛋白的表達(dá),并比較三種蛋白在這兩種癌組織和正常眼瞼組織中的表達(dá)差異和在不同分期的眼瞼鱗癌組織中的表達(dá)差異,X2檢驗(yàn)比較各因子表達(dá)陽性率的差異。觀察眼瞼鱗癌和基底細(xì)胞癌中AP-1、 PDCD4與p73蛋白表達(dá)與性別、年齡、病理分期等的相互關(guān)系。 結(jié)果 1AP-1在眼瞼鱗癌和基底細(xì)胞癌中陽性表達(dá)率明顯高于正常的眼瞼組織,經(jīng)統(tǒng)計學(xué)處理,差異有顯著性(P0.05)。鱗癌和基底細(xì)胞癌中的AP-1陽性表達(dá)率無顯著差異(P0.05)。AP-1在眼瞼鱗癌和基底細(xì)胞癌患者中的表達(dá)與年齡、性別、有無淋巴轉(zhuǎn)移無顯著性差異;而在鱗癌不同的病理分期中的陽性表達(dá)率差異有統(tǒng)計學(xué)意義(P0.05)。 2PDCD4蛋白在眼瞼正常組織中高表達(dá),而在鱗癌和基底細(xì)胞癌中陽性表達(dá)率明顯低于正常的眼瞼組織,經(jīng)統(tǒng)計學(xué)處理,差異有顯著性(P0.05)。鱗癌和基底細(xì)胞癌中的PDCD4陽性表達(dá)率無顯著差異(P0.05)。PDCD4在鱗癌和基底細(xì)胞癌患者中的表達(dá)與年齡、性別、有無淋巴轉(zhuǎn)移無顯著性差異;而在鱗癌不同的病理分期中的陽性表達(dá)率差異有統(tǒng)計學(xué)意義(P0.05)。 3p73在正常眼瞼組織的基底細(xì)胞層表達(dá),而隨著細(xì)胞的角化,p73表達(dá)下降。鱗癌中p73表達(dá)輕度升高,與眼瞼正常組織差異無統(tǒng)計學(xué)意義(P0.05),基底細(xì)胞癌中p73表達(dá)明顯升高,陽性表達(dá)率有統(tǒng)計學(xué)差異(P0.05)。p73在鱗癌和基底癌患者中的表達(dá)與年齡、性別無顯著性差異;而在鱗癌不同的病理分期中的陽性表達(dá)率差異具統(tǒng)計學(xué)意義(P0.05)。 4AP-1與PDCD4兩種蛋白在鱗癌中的表達(dá)呈負(fù)相關(guān),r=-0.401(P0.05),AP-1與p73,p73與PDCD4兩組蛋白在鱗癌中的表達(dá)無相關(guān)性(P0.05)。 結(jié)論 1AP-1蛋白的陽性表達(dá)率在鱗癌和基底癌組織中顯著高于正常眼瞼組織,AP-1蛋白在不同的鱗癌病理分期中的陽性表達(dá)率有顯著性差異,提示AP-1與鱗癌的發(fā)生、發(fā)展密切相關(guān)。 2PDCD4在正常眼瞼組織中表達(dá)率高于眼瞼鱗癌和基底細(xì)胞癌,差異有統(tǒng)計學(xué)意義,提示PDCD4與鱗癌和基底細(xì)胞癌的發(fā)生發(fā)展可能成負(fù)相關(guān),PDCD4可能在某種程度上抑制眼瞼癌癥的發(fā)生與發(fā)展。 3p73在正常眼瞼組織的基底細(xì)胞層高表達(dá),而隨著細(xì)胞的角化,p73表達(dá)下降。鱗癌中p73表達(dá)輕度升高,差異無統(tǒng)計學(xué)意義;基底細(xì)胞癌中p73表達(dá)升高,差異具有統(tǒng)計學(xué)意義,提示p73可能與細(xì)胞的分化相關(guān)。 4AP-1與PDCD4在眼瞼鱗癌中表達(dá)呈負(fù)相關(guān),提示AP-1可能與PDCD4蛋白在參與眼瞼鱗癌發(fā)生中存在某種相互關(guān)系。
[Abstract]:Objective To observe the expression and clinical significance of AP-1 (activation protein -1), PDCD4 (programmed cell death factor -4) and p73 protein in different pathological stages and basal cell carcinoma of eyelid squamous cell carcinoma, and to explore the relationship between the three and the occurrence and development of eyelid basal cell carcinoma and squamous cell carcinoma.
Methods: 68 cases of eyelid squamous cell carcinoma and 40 cases of basal cell carcinoma were detected by immunohistochemical staining. The expression of AP-1, PDCD4 and p73 in the three tissues was detected in 20 normal eyelid tissues, and the differences in the expression of three proteins in the two kinds of cancerous tissues and normal eyelid tissues and the eyelid scales in different stages were compared. The difference of expression in cancer tissues. X2 test compared the difference of the positive rates of various factors. The relationship between the expression of AP-1, PDCD4 and p73 protein in the eyelid squamous cell carcinoma and the basal cell carcinoma was observed with sex, age and pathological stage.
Result
The positive expression rate of 1AP-1 in eyelid squamous cell carcinoma and basal cell carcinoma was significantly higher than that of normal eyelid tissue. The difference was significant (P0.05). There was no significant difference in the positive rate of AP-1 in squamous and basal cell carcinoma (P0.05). The expression of.AP-1 in eyelid squamous cell carcinoma and basal cell carcinoma was associated with age, sex, and lymph node metastasis. There was no significant difference, but there was a significant difference in the positive expression rate in different pathological stages of squamous cell carcinoma (P0.05).
The expression of 2PDCD4 protein was highly expressed in normal eyelid tissue, while the positive expression rate in squamous and basal cell carcinoma was significantly lower than that in normal eyelid tissue. The difference was significant (P0.05). There was no significant difference in the positive rate of PDCD4 in squamous and basal cell carcinoma (P0.05) and the expression of.PDCD4 in squamous and basal cell carcinoma patients There was no significant difference in age, sex and lymph node metastasis, but there was a significant difference in the positive expression rate in different pathological stages of squamous cell carcinoma (P0.05).
The expression of 3p73 was expressed in the basal cell layer of normal eyelid tissue, and the expression of p73 decreased with the keratinization of the cells. The expression of p73 in the squamous cell carcinoma was slightly elevated, and there was no statistical difference between the normal tissue and the normal eyelid (P0.05). There was a negative correlation between the expression of 4AP-1 and PDCD4 protein in squamous cell carcinoma, r=-0.401 (P0.05), and there was no correlation between AP-1 and p73, p73 and PDCD4 two histone expression in squamous cell carcinoma (P0.05). Two.
conclusion
The positive expression rate of 1AP-1 protein was significantly higher in the squamous and basal tissues than in the normal eyelid tissue. The positive expression rate of AP-1 protein in the pathological stages of different squamous cell carcinoma showed significant difference, suggesting that AP-1 was closely related to the development of squamous cell carcinoma.
The expression rate of 2PDCD4 in normal eyelid tissue is higher than that of eyelid squamous cell carcinoma and basal cell carcinoma. The difference is statistically significant, suggesting that PDCD4 may be negatively related to the development and development of squamous cell carcinoma and basal cell carcinoma. PDCD4 may inhibit the occurrence and development of eyelid cancer to some extent.
