臨床喉鱗癌組織標本中G3BP和CD44v6的檢測及意義

【摘要】:目的研究RNA結合的Ras-GAP酶激活蛋白SH3結合蛋白(G3BP)、腫瘤干細胞標志物CD44v6蛋白與喉鱗狀細胞癌(喉鱗癌)發(fā)生發(fā)展的關系,探討G3BP、CD44v6蛋白與血管新生的相關性。方法收集56例喉鱗癌患者癌組織及對應癌旁組織,采用Western blot法檢測喉鱗癌患者癌組織及對應癌旁組織中G3BP、CD44v6蛋白水平,使用免疫組織化學染色法分別檢測G3BP、CD44v6、血管內(nèi)皮生長因子A(VEGF-A)的表達,并分析G3BP、CD44v6表達與喉鱗癌患者臨床病理特征的關系及與VEGF-A表達的相關性;采用Ⅷ因子相關抗原(FⅧAg)免疫組織化學染色確定微血管密度(MVD),觀察G3BP、CD44v6表達與MVD之間的關系。結果喉鱗癌患者癌組織中G3BP蛋白、CD44v6蛋白水平均明顯高于對應癌旁的正常組織。喉鱗癌患者癌組織中的G3BP、CD44v6、VEGF-A表達陽性率分別為58.9%、53.6%、46.4%,均明顯高于對應的癌旁正常組織。喉鱗癌患者癌組織中G3BP、CD44v6蛋白表達陽性率與臨床分期、復發(fā)或轉移及淋巴結轉移相關,但與年齡和腫瘤大小無關。經(jīng)Pearson相關性分析,喉鱗癌患者癌組織中G3BP、CD44v6蛋白的表達與VEGF-A表達呈正相關(r=0.741,r=0.756);喉鱗癌患者癌組織中G3BP與CD44v6蛋白陽性表達病例的MVD值均明顯高于癌旁正常組織中MVD值;癌組織中G3BP與CD44v6蛋白陰性表達病例MVD值與癌旁正常組織中MVD值比較無明顯差異。結論喉鱗癌組織G3BP、CD44v6蛋白高表達,可能通過促進VEGF-A的表達來誘發(fā)腫瘤微血管的新生,促進喉鱗癌侵襲、轉移。
[Abstract]:Objective to study the relationship between RNA binding Ras-GAP activator protein SH3 binding protein G3BP1, tumor stem cell marker CD44v6 protein and laryngeal squamous cell carcinoma (LSCC), and to explore the relationship between G3BP- CD44v6 protein and angiogenesis in laryngeal squamous cell carcinoma (LSCC). Methods A total of 56 cases of laryngeal squamous cell carcinoma (LSCC) and its adjacent tissues were collected. The levels of G3BP- CD44v6 protein in laryngeal squamous cell carcinoma (LSCC) tissues and adjacent tissues were detected by Western blot assay.
