【摘要】：目的運用Tet-on 3G四環(huán)素誘導表達系統(tǒng)探討β-神經(jīng)生長因子(β-NGF)在人胚腎(HEK293FT)細胞中的過表達情況及其對角膜緣干細胞(LSCs)的作用。方法將p LVX-TRE3G-IRES-β-NGF和p LVX-Tet3G慢病毒在空白的HEK293FT細胞進行擴增,收獲慢病毒,再共轉(zhuǎn)染HEK293FT細胞,同時用不同劑量強力霉素(Dox)誘導β-NGF表達(分為未轉(zhuǎn)染組、1000、100和1000μg/L Dox誘導表達組)。48 h后收集細胞,免疫印跡法(Western blotting)檢測各組細胞內(nèi)β-NGF表達情況。體外分離培養(yǎng)LSCs,慢病毒共轉(zhuǎn)染LSCs,分為對照組(未轉(zhuǎn)染組)和誘導表達組[實驗組A(慢病毒載體Dox 1000μg/L)、實驗組B(慢病毒載體Dox 100μg/L)、實驗組C(慢病毒空載體Dox 1000μg/L)],誘導表達48 h后細胞免疫熒光染色檢測NGF及相關蛋白(p63、p38、Trk A)的表達情況。結(jié)果NGF在轉(zhuǎn)染細胞內(nèi)被誘導表達,隨著Dox劑量增加,表達量增強,差異有統(tǒng)計學意義(P0.05)。與對照組及實驗組C相比,實驗組A的p63表達降低,Trk A表達降低,p38表達增加,差異有統(tǒng)計學意義(P0.05);與對照組及實驗組C相比,實驗組B的p63表達增加,Trk A表達增加,p38表達降低,差異有統(tǒng)計學意義(P0.05)。結(jié)論運用Tet-on 3G四環(huán)素誘導表達系統(tǒng),可成功誘導表達不同劑量β-NGF蛋白,低劑量Dox誘導下NGF產(chǎn)生的微環(huán)境可促進LSCs的體外擴增,并能維持LSCs的干細胞特性。
[Abstract]:Objective to investigate the overexpression of 尾 -NGF in human embryonic kidney cell line HEK293FTs and its effect on limbal stem cells by using Tet-on 3G tetracycline induced expression system. Methods pLVX-TRE3G-IRES- 尾 -NGF and p LVX-Tet3G lentivirus were amplified in blank HEK293FT cells. Lentivirus was harvested and co-transfected into HEK293FT cells. 尾 -NGF expression was induced by doxycycline in different doses. The expression of 尾 -NGF was detected by Western blotting. LSCswere isolated and cultured in vitro. Lentivirus co-transfected LSCswere divided into two groups: control group (untransfected group) and induced expression group [experimental group A (lentivirus vector Dox 1000 渭 g / L), experimental group B (lentivirus). The expression of NGF and its associated protein, p63, p38, Trk A, was detected by cell immunofluorescence staining 48 h after induction and expression of lentivirus empty vector Dox (1000 渭 g / L). Results the expression of NGF was induced in transfected cells. Compared with the control group and the experimental group C, the expression of p63 in experimental group A decreased and the expression of TrkA decreased, and the expression of p38 increased. Compared with the control group and the experimental group C, the expression of p63 in experimental group B increased, and the expression of p38 decreased. Conclusion the expression system of tetracycline induced by Tet-on 3G was used. 尾 -NGF protein was successfully induced and expressed in different doses. The microenvironment of NGF induced by low dose Dox could promote the expansion of LSCs in vitro and maintain the stem cell character of LSCs.
