注射神經(jīng)生長因子對(duì)兔眼外肌的影響

【摘要】:目的觀察大白兔眼外肌注射神經(jīng)生長因子(NGF)對(duì)眼外肌的影響。方法將8只大白兔右眼設(shè)為觀察組,左眼設(shè)為對(duì)照組,觀察組眼上直肌注射10μg NGF;對(duì)照組眼上直肌相同位置注射等量生理鹽水。測(cè)量眼外肌濕重,計(jì)算眼外肌橫截面積,HE染色測(cè)量肌纖維周徑、觀察炎癥情況,免疫組化染色測(cè)定活化衛(wèi)星細(xì)胞。結(jié)果觀察組上直肌橫截面積[(12.412±1.372)mm2]顯著大于對(duì)照組[(6.738±0.856)mm~2,P0.05]。觀察組兔肌纖維周徑[(228.32±16.57)μm]顯著大于對(duì)照組[(129.32±11.16)μm,P0.05]。觀察組大、中、小細(xì)胞構(gòu)成比分別為27.0%、69.6%、3.5%,對(duì)照組大、中、小細(xì)胞構(gòu)成比分別為13.4%、63.5%、23.2%,兩組比較差異有統(tǒng)計(jì)學(xué)意義(P0.05)。HE染色:治療后2 w,兩組兔眼外肌均沒有炎癥細(xì)胞浸潤和細(xì)胞壞死,肌纖維邊界清楚、方向一致、排列整齊。免疫組化染色:觀察組活化衛(wèi)星細(xì)胞百分率[(28.78±4.36)%]顯著大于對(duì)照組[(11.87±2.47)%,P0.05]。結(jié)論眼外肌注射NGF能夠活化衛(wèi)星細(xì)胞,促進(jìn)眼外肌肌纖維增生,對(duì)眼組織沒有明顯不良影響。
[Abstract]:Objective to observe the effect of nerve growth factor (NGF) injection into the extraocular muscles of rabbits. In the observation group, the superior rectus muscle was injected with 10 渭 g NGF and the control group was injected with the same amount of normal saline at the same position. The wet weight of the extraocular muscle was measured, the cross sectional area of the extraocular muscle was calculated and the diameter of the muscle fiber was measured by HE staining, and the inflammation was observed. Results the cross sectional area of the superior rectus muscle in the observation group [12.412 鹵1.372)mm2] was significantly larger than that in the control group [6.738 鹵0.856 mm]. The peripheral diameter of muscle fiber in the observation group [228.32 鹵16.57 渭 m] was significantly higher than that in the control group [129.32 鹵11.16 渭 m P0.05]. The ratio of large, middle and small cells in the observation group was 27.00.69.63.5 渭 m, and that in the control group was larger than that in the control group. The ratio of small cells to small cells was 13. 4% and 63. 5% and 23. 2, respectively. The difference between the two groups was statistically significant (P 0. 05). He staining: at 2 weeks after treatment, there were no inflammatory cell infiltration and cell necrosis in the extraocular muscles of the two groups, and the muscle fiber boundary was clear and the direction was the same. Immunohistochemical staining: the percentage of activated satellite cells in the observation group [28.78 鹵4.36] was significantly higher than that in the control group [11.87 鹵2.47]. Conclusion injection of NGF into the extraocular muscle can activate the satellite cells, promote the proliferation of the muscle fiber of the extraocular muscle, and have no obvious adverse effect on the ocular tissue.
