角膜新生血管和淋巴管與房水中VEGF-A和VEGF-C表達(dá)量關(guān)系的臨床研究

【摘要】:目的:通過對因角膜病變行角膜移植患者的角膜組織中的角膜新生血管(Corneal neovascularization,CNV)和角膜新生淋巴管(Corneallymphangiogenesis,CL)行免疫組化染色計數(shù)以及檢測其房水中血管內(nèi)皮生長因子A(Vascular endothelial growth factor-A,VEGF-A)和血管內(nèi)皮生長因子C(Vascular endothelial growth factor-C,VEGF-C)的表達(dá)的研究,建立房水中VEGF-A和VEGF-C與角膜新生血管和角膜新生淋巴管之間的定量關(guān)系,揭示房水中VEGF-A和VEGF-C表達(dá)量與角膜新生血管和角膜新生淋巴管數(shù)變化規(guī)律,,為角膜病患者實行角膜移植的手術(shù)時機(jī)選擇提供理論依據(jù)。 方法:10例因各類角膜潰瘍和堿燒傷后需要行角膜移植的患者納入本研究,根據(jù)病史記錄病程長短,以及術(shù)前患者的裂隙燈照片,了解患者術(shù)前情況。采用管腔內(nèi)皮非特異受體CD31,定量檢測角膜新生血管數(shù)目(Blood vessels counting,BVC),采用淋巴管內(nèi)皮細(xì)胞特異性抗體LYVE-1行免疫組織化學(xué)染色,定量檢測角膜新生淋巴管的數(shù)目(Lymphatic vessels counting, LVC)。將10個實驗組樣本和10個對照組(單純年齡相關(guān)性白內(nèi)障患者)的房水用酶聯(lián)免疫吸附測定法(Enzyme linked immunosorbentassay,ELISA)對比試驗,測出房水中VEGF-A和VEGF-C的表達(dá)量。探索房水中VEGF-A和VEGF-C的表達(dá)量與BVC和LVC之間的關(guān)系。 結(jié)果:免疫組化染色結(jié)果10例實驗組角膜都有新生血管,4例既有新生血管又有新生淋巴管。實驗組和對照組房水中VEGF-A和VEGF-C表達(dá)量有顯著性差異,角膜新生血管、新生淋巴管與其房水的VEGF-A和VEGF-C的表達(dá)量有明顯相關(guān)性。 結(jié)論:角膜病變后有角膜新生血管和新生淋巴管的患者,其房水中的VEGF-A和VEGF-C的表達(dá)量明顯增高,且VEGF-A和角膜新生血管的數(shù)量呈正相關(guān),VEGF-C和角膜新生淋巴管歸一化后變化趨勢分析提示其同步性非常高,故通過房水中VEGF-A和VEGF-C的檢測可推知角膜新生血管和淋巴管的生長情況。
[Abstract]:Objective: to count the corneal neovascularization (endothelial) and the corneal neovascularization (endothelial) in corneal tissue of corneal transplant patients with keratoplasty due to keratopathy, and to detect A(Vascular endothelial growth in aqueous humor by immunohistochemical staining. The expression of VEGF-A and C(Vascular endothelial growth factor-C- VEGF-C) was studied. To establish a quantitative relationship between VEGF-A and VEGF-C in aqueous humor and corneal neovascularization and corneal neovascularization lymphatic vessels, and to reveal the changes of VEGF-A and VEGF-C expression in aqueous humor and the number of corneal neovascularization and corneal neovascularization. To provide a theoretical basis for the choice of surgical timing for corneal transplantation in patients with keratopathy. Methods Ten patients who needed corneal transplantation due to various corneal ulcers and alkali burns were included in the study. The duration of the disease was recorded according to the history and the slit lamp photos of the patients before operation. To understand the preoperative status of the patients, the endothelial nonspecific receptor CD31 was used to quantitatively detect the number of corneal neovascularization and the number of blood vessels counting BVCs. The lymphatic endothelial cell-specific antibody (LYVE-1) was used for immunohistochemical staining. Quantitative detection of the number of lymphatic vessels in cornea (Lymphatic vessels counting). The aqueous humor of 10 experimental group samples and 10 control groups (simple age-related cataract patients) were tested by enzyme linked immunosorbent immunosorbent assay (Elisa) and compared with that of 10 control groups (simple age-related cataract patients) by enzyme-linked immunosorbent immunosorbent assay (Elisa). The expression of VEGF-A and VEGF-C in aqueous humor was measured and the relationship between the expression of VEGF-A and VEGF-C and BVC and LVC in aqueous humor was explored. Results: the expression of VEGF-A and VEGF-C in aqueous humor was significantly different between experimental group and control group, and the corneal neovascularization was found in 4 cases with neovascularization and lymphatic vessels, and the expression of VEGF-A and VEGF-C in aqueous humor of experimental group and control group were significantly different. There was a significant correlation between the expression of VEGF-A and VEGF-C in aqueous humor. Conclusion: the expression of VEGF-A and VEGF-C in aqueous humor of patients with corneal neovascularization and neovascularization after keratopathy was significantly increased. VEGF-A and corneal neovascularization were positively correlated with the number of corneal neovascularization. The trend analysis of the changes after normalization of corneal neovascularization showed that their synchronism was very high. Therefore, the growth of corneal neovascularization and lymphatic vessels could be inferred by the detection of VEGF-A and VEGF-C in aqueous humor.
