脈沖噪聲暴露后大鼠下丘核生長相關蛋白-43表達的研究

【摘要】:目的探討脈沖噪聲暴露對大鼠下丘核生長相關蛋白-43(growth associated protein,Gap-43)表達的影響。方法 SPF級成年雄性SD大鼠30只,隨機分為正常對照組6只、噪聲暴露組24只,正常對照組不接受噪聲暴露,噪聲暴露組接受脈沖噪聲暴露,噪聲平均壓力峰值156dB SPL,脈寬0.25ms,單次暴露6s,共暴露50次。于噪聲暴露前及暴露后3、7、14、28d分別測試對照組及噪聲暴露組(每個時間點6只)大鼠聽性腦干反應(auditory brainstem response,ABR)閾值;然后取雙側下丘進行免疫組化染色,通過灰度值檢測各組大鼠下丘Gap-43蛋白表達水平。結果噪聲暴露后各組大鼠ABR反應閾顯著高于正常組(P0.01),噪聲暴露后14、28d組ABR閾值顯著低于暴露后3d組(P0.05);噪聲暴露后各組大鼠雙側下丘Gap-43蛋白表達顯著高于正常組(P0.01),暴露后28d組大鼠下丘Gap-43蛋白表達顯著低于暴露后3d組(P0.05)。結論脈沖噪聲暴露可導致大鼠ABR反應閾顯著升高,下丘Gap-43蛋白高表達;脈沖噪聲暴露后不同時間大鼠ABR閾值升高和下丘Gap-43蛋白高表達的變化規(guī)律可能與聽皮層突觸重塑有關。
[Abstract]:Objective to investigate the effect of pulsed noise exposure on the expression of Gap-43 associated protein associated protein in the inferior colliculus nucleus of rats. Methods Thirty adult male SD rats of SPF grade were randomly divided into normal control group (n = 6) and noise exposure group (n = 24). The normal control group did not receive noise exposure, while the noise exposure group received impulse noise exposure. The mean noise pressure peak value was 156dB SPLL, pulse width 0.25ms, single exposure 6s, exposure 50 times. The auditory brainstem response (ABR) thresholds of the control group and the noise exposure group (6 rats at each time point) were measured before and after exposure to noise for 3 weeks and 14 days after exposure, and the threshold values of auditory brainstem response (ABR) were measured respectively in the control group and the noise exposure group (6 rats at each time point). Then the bilateral inferior colliculus was taken for immunohistochemical staining. Results the ABR response threshold of rats after noise exposure was significantly higher than that of normal group (P 0.01), and the threshold of ABR in group 14: 28 days after noise exposure was significantly lower than that in group 3 days after exposure to noise (P 0.05). The expression of Gap-43 protein in the bilateral inferior colliculus of the rats in the control group was significantly higher than that in the control group (P 0.01), and the expression of Gap-43 protein in the inferior colliculus of the rats 28 days after exposure was significantly lower than that in the group of 3 days after exposure. Conclusion Pulsed noise exposure can significantly increase the threshold of ABR response in rats. The high expression of Gap-43 protein in inferior colliculus and the increase of ABR threshold and Gap-43 protein expression in inferior colliculus of rats at different time after impulse noise exposure may be related to synaptic remodeling of auditory cortex.
