局部注射鼠神經(jīng)生長因子對眼外肌的作用研究

【摘要】:目的探討兔眼外肌局部注射鼠神經(jīng)生長因子(nerve growth factor, NGF)后的肌肉收縮能力及組織學(xué)變化,探索藥物治療斜視的新方法。 方法以16只成年新西蘭大白兔為實(shí)驗(yàn)對象,左眼上直肌注射0.05mL(5μg) NGF(實(shí)驗(yàn)組),右眼上直肌注射0.05mL生理鹽水(對照組)。1周后將實(shí)驗(yàn)動物麻醉測量上直肌肌肉收縮力,測量完畢處死動物后獲取上直肌,測量肌肉濕重,長度,計(jì)算肌肉橫截面積,做石蠟切片并行形態(tài)學(xué)檢查。 結(jié)果(1)實(shí)驗(yàn)組眼外肌在單刺激及不同頻率刺激下,肌力較對照組均明顯增強(qiáng),差異均有統(tǒng)計(jì)學(xué)意義(P0.05)。實(shí)驗(yàn)組上直肌抗疲勞性實(shí)驗(yàn)結(jié)果(166.00±12.46)s較對照組(150.30±14.49)s差異有統(tǒng)計(jì)學(xué)意義(P)0.05)。(2)兩組肌肉濕重未見明顯不同,實(shí)驗(yàn)組眼外肌肌肉橫截面面積為(9.67±1.82)mm2,對照組眼外肌肌肉橫截面面積為(6.97±1.90)mm2,實(shí)驗(yàn)組較對照組增大明顯,差異有統(tǒng)計(jì)學(xué)意義(P0.05)。注射部位無明顯炎性細(xì)胞浸潤,未見眼外肌肌纖維變性、壞死等毒性反應(yīng)。注射部位附近的鞏膜和相應(yīng)的色素膜也未見明顯炎性變化。Van Gieson氏苦味酸酸性復(fù)紅染色未見膠原異常增生。實(shí)驗(yàn)組眼外肌纖維周徑為(167.20±35.55)μm,較對照組(127.44±33.64)μ m更大(P0.01)。實(shí)驗(yàn)組眼外肌纖維大細(xì)胞計(jì)數(shù)為136、中等細(xì)胞計(jì)數(shù)為381、小細(xì)胞計(jì)數(shù)為28,對照組分別為88、383、118,實(shí)驗(yàn)組大細(xì)胞和中等細(xì)胞比例較對照組增高,小細(xì)胞比例降低,差異有統(tǒng)計(jì)學(xué)意義(P0.01)。(3)實(shí)驗(yàn)組PCNA陽性細(xì)胞百分率為(21.93±5.08)%較對照組(12.97±2.68)%更大,差異有統(tǒng)計(jì)學(xué)意義(P0.01)。實(shí)驗(yàn)組PCNA(?)日性細(xì)胞總數(shù)占總細(xì)胞數(shù)的21.8%(664/3078),對照組為12.1%(347/2869),差異有統(tǒng)計(jì)學(xué)意義(P0.01)。 結(jié)論眼外肌局部注射5μg鼠神經(jīng)生長因子對眼部組織無明顯不良作用。眼外肌局部注射鼠神經(jīng)生長因子能短期迅速提高肌肉收縮能力。鼠神經(jīng)生長因子能促使眼外肌肌纖維快速增生,并能促進(jìn)組織細(xì)胞分化。
[Abstract]:Objective to investigate the muscle contractility and histological changes after local injection of nerve growth factor nerve growth factor (NGF) into rabbit extraocular muscles, and to explore a new method for the treatment of strabismus. Methods Sixteen adult New Zealand white rabbits were used to measure the contractility of the superior rectus muscle by injecting 0.05mLL of 5 渭 g of NGF into the left superior rectus muscle (experimental group) and 0.05 mL saline (the control group was injected with 0.05 mL normal saline) after 1 week of anaesthesia, and the contractility of the muscle of the superior rectus muscle was measured. After the animals were killed, the superior rectus muscle was obtained, the wet weight and length of the muscle were measured, the cross sectional area of the muscle was calculated, the paraffin section was made and the morphological examination was performed. Results 1) the muscle strength of the extraocular muscle in the experimental group was significantly higher than that in the control group under single stimulation and different frequency stimulation. There was no significant difference in muscle wet weight between the experimental group and the control group (166.00 鹵12.46 vs 150.30 鹵14.49 vs 150.30 鹵14.49). The cross sectional area of the extraocular muscle in the experimental group was 9.67 鹵1.82 mm ~ (-2), and that in the control group was 6.97 鹵1.90 mm ~ (2), which was significantly larger than that in the control group (P < 0.05). There was no significant infiltration of inflammatory cells and no degeneration of the extraocular muscle at the injection site. No inflammatory changes were observed in the sclera near the injection site and the corresponding pigment membrane. There was no abnormal proliferation of collagen by van Gieson picric acid acid fuchsin staining. The peripheral diameter of the extraocular muscle fibers in the experimental group was 167.20 鹵35.55 渭 m, which was higher than that in the control group. In the experimental group, the number of fibrous large cells in extraocular muscle was 136, the number of medium cells was 381, the number of small cells was 28, and the number of small cells in the control group was 88-383118.The ratio of large cells and medium cells in the experimental group was higher than that in the control group. The percentage of PCNA positive cells in the experimental group was 21.93 鹵5.08%, which was higher than that in the control group (12.97 鹵2.68%). ) the total number of diurnal cells accounted for 21.8% of the total number of cells, and that of the control group was 12.1%, 347 / 2869% (P 0.01). Conclusion Local injection of 5 渭 g nerve growth factor into the extraocular muscle has no obvious adverse effect on ocular tissue. Local injection of nerve growth factor into the extraocular muscle can improve the contractility of the muscle in a short period of time, and nerve growth factor can promote the eye. The muscle fibers of the external muscles proliferate rapidly. And can promote tissue and cell differentiation.
