Snail、E-cadherin、VEGF蛋白及其mRNA在喉鱗癌中的表達(dá)研究

【摘要】:本課題通過應(yīng)用EliVisionTM Plus免疫組織化學(xué)技術(shù)(Immunohistochemistry, IHC)(?)原位分子雜交技術(shù)(In situ hybridization,ISH)分別檢測(cè)喉黏膜慢性炎、不典型增生、鱗癌(Laryngeal squamous cell carcinoma, LSCC)組織中Snail、E-cadherin和VEGF蛋白及其mRNA的表達(dá)情況。 探討Snail、E-cadherin和VEGF蛋白及其mRNA分別在喉黏膜慢性炎、不典型增生和鱗癌組織中的表達(dá)及與患者臨床病理特征的關(guān)系;探討Snail、E-cadherin和VEGF蛋白在喉鱗癌組織中表達(dá)的相互關(guān)系;探討Snail、E-cadherin和VEGF mRNA在喉鱗癌組織中表達(dá)的相互關(guān)系;免疫組織化學(xué)技術(shù)和原位分子雜交技術(shù)兩種方法檢測(cè)喉鱗癌組織中Snail、E-cadherin和VEGF表達(dá)的一致性分析。 收集解放軍第251醫(yī)院病理科2005年1月至2007年1月間的喉鱗癌術(shù)后標(biāo)本60例、不典型增生30例和黏膜慢性炎組織20例,分別采用EliVisionTM Plus免疫組織化學(xué)技術(shù)和原位分子雜交技術(shù)檢測(cè)Snail、E-cadherin和VEGF蛋白及其mRNA的表達(dá)情況,并分析它們與患者的性別、年齡、病理分級(jí)、TNM分期、T分期、甲狀軟骨累及、淋巴結(jié)轉(zhuǎn)移及遠(yuǎn)處轉(zhuǎn)移的關(guān)系;分析兩種方法檢測(cè)Snail、E-cadherin和VEGF表達(dá)的一致性。 Snail、E-cadherin和VEGF蛋白及其mRNA在60例喉鱗癌組織中的陽性率分別為:78.3%(47/60)、46.7%(28/60)、80.0%(48/60)及70.0%(42/60)、45.0%(27/60)、73.3%(44/60);在30例喉黏膜不典型增生組織中的陽性率分別為:43.3%(13/30)、76.7%(23/30)、36.7%(11/30)及20.0%(6/30)、70.0%(21/30)、26.7%(8/30);在20例喉黏膜慢性炎組織中的陽性率分別為:15.0%(3/20)、100%(20/20)、15.0%(3/20)及5.0%(1/20)、90.0%(18/20)、5.0%(1/20)。Snail、VEGF蛋白及其mRNA在喉鱗癌組織的陽性率顯著高于不典型增生和黏膜慢性炎組織的陽性率,E-cadherin蛋白及其mRNA在喉鱗癌組織的陽性率均顯著低于不典型增生和黏膜慢性炎組織的陽性率,差異具有統(tǒng)計(jì)學(xué)意義。不典型增生組織中Snail蛋白陽性率高于黏膜慢性炎組織的表達(dá)率,差異有統(tǒng)計(jì)學(xué)意義,Snail mRNA、 VEGF蛋白及其mRNA陽性率高于黏膜慢性炎組織的表達(dá)率,但差異無統(tǒng)計(jì)學(xué)意義。不典型增生組織中E-cadherin蛋白陽性率低于黏膜慢性炎組織的表達(dá)率,差異具有統(tǒng)計(jì)學(xué)意義,E-cadherin mRNA陽性率低于黏膜慢性炎組織的表達(dá)率,但差異無統(tǒng)計(jì)學(xué)意義;Snail、 VEGF蛋白及mRNA在喉鱗癌組織中的高表達(dá)及其E-cadherin蛋白及mRNA的低表達(dá)與腫瘤TNM分期、T分期、甲狀軟骨侵犯、淋巴結(jié)轉(zhuǎn)移、遠(yuǎn)處轉(zhuǎn)移及5年生存率密切相關(guān),而與年齡、性別無明顯關(guān)系;Snail蛋白表達(dá)與病理分級(jí)有關(guān),而Snail mRNA的表達(dá)與病理分級(jí)無關(guān)。 60例喉鱗癌組織中Snail、E-cadherin、 VEGF蛋白及mRNA陽性表達(dá)分別為47例、28例、48例及42例、27例、44例,經(jīng)Spearman相關(guān)分析,結(jié)果顯示:Snail蛋白及mRNA與E-cadherin蛋白及mRNA表達(dá)呈顯著負(fù)相關(guān);Snail蛋白及mRNA與VEGF蛋白及mRNA表達(dá)呈顯著正相關(guān);E-cadherin蛋白及mRNA與VEGF蛋白及mRNA表達(dá)呈顯著負(fù)相關(guān)。 Snail過表達(dá)與喉鱗癌的發(fā)生有密切關(guān)系,它可能參與了癌前細(xì)胞向癌細(xì)胞轉(zhuǎn)化的調(diào)控機(jī)制;Snail、 E-cadherin和VEGF在免疫組化和原位雜交兩種檢測(cè)方法中具有一致性;Snail、 E-cadherin和VEGF共同參與了喉鱗癌的發(fā)生、發(fā)展、浸潤(rùn)、轉(zhuǎn)移過程,是喉鱗癌發(fā)生浸潤(rùn)和轉(zhuǎn)移的重要生物學(xué)指標(biāo),聯(lián)合檢測(cè)Snail、 E-cadherin和VEGF對(duì)預(yù)測(cè)腫瘤轉(zhuǎn)移和預(yù)后有重要意義,Snail將有可能成為喉鱗癌靶向治療的一個(gè)新指標(biāo)。
[Abstract]:This topic through the application of EliVisionTM Plus immunohistochemical technique (Immunohistochemistry, IHC) (?) in situ hybridization (In situ hybridization, ISH) were detected in laryngeal mucosa of chronic inflammation, atypical hyperplasia, squamous cell carcinoma (Laryngeal squamous cell carcinoma, LSCC) in Snail, the expression of E-cadherin and VEGF protein and mRNA.
The effects of Snail, E-cadherin and VEGF protein and mRNA respectively in laryngeal mucosa of chronic inflammation, the expression of atypical hyperplasia and squamous cell carcinoma and its relationship with the clinical pathological features of patients; to investigate the Snail, relationship between expression of E-cadherin and VEGF protein in laryngeal squamous cell carcinoma; to explore the relationship between the expression of Snail, E-cadherin and VEGF mRNA in laryngeal squamous cell carcinoma the organization of Snail in laryngeal squamous cell carcinoma; immunohistochemistry and in situ hybridization of two kinds of detection methods, consistency analysis of the expression of E-cadherin and VEGF.
60 samples were collected from the 251st Hospital of PLA from January 2005 to January 2007 in the Department of pathology of postoperative laryngeal squamous cell carcinoma, 30 cases of atypical hyperplasia and chronic inflammation of mucosa tissues in 20 cases were measured by Snail EliVisionTM Plus immunohistochemistry and in situ hybridization, the expression of E-cadherin and VEGF protein and mRNA, and analyze them with patients the gender, age, pathological grading, TNM staging, T staging, thyroid cartilage involvement, lymph node metastasis and distant metastasis; analysis of two methods for detection of Snail, E-cadherin and VEGF expression of consistency.
