PPAR-γ在甲狀腺相關(guān)性眼病患者眶脂肪和血清中的表達(dá)研究

【摘要】:目的:檢測(cè)TAO患者眼眶脂肪組織以及血清中PPAR-γ的表達(dá)水平,初步探討PPAR-γ在TAO發(fā)病過程中的作用。方法:選取TAO患者4例,健康對(duì)照者4例,用RT-PCR法檢測(cè)兩組眶脂肪PPAR-γmRNA的表達(dá)水平以及用Western blot法檢測(cè)兩組眶脂肪PPAR-γ蛋白的表達(dá)水平。選取TAO患者33例(男14例,女19例),按臨床活動(dòng)性評(píng)分(CAS)及99Tcm-DTPA眼眶顯像將TAO患者分為兩大組:活動(dòng)期(CAS≥3,99Tcm-DTPA眼眶顯像陽性)18例(男7例,女11例)和非活動(dòng)期(CAS3,99Tcm-DTPA眼眶顯像陰性)15例(男7例,女8例)。單純Graves病(GD)患者16例(男6例,女10例),健康對(duì)照者(CON)15例(男7例,女8例)。用Elisa法檢測(cè)四組血清PPAR-γ表達(dá)水平。采用SPSS19.0進(jìn)行統(tǒng)計(jì)分析,計(jì)量資料采用x±s表示,計(jì)數(shù)資料采用例數(shù)(構(gòu)成比)表示,正態(tài)分布數(shù)據(jù)均數(shù)的比較采用方差分析,非正態(tài)分布數(shù)據(jù)均數(shù)的比較采用秩和檢驗(yàn),構(gòu)成比的比較采用卡方檢驗(yàn),檢驗(yàn)水準(zhǔn)α=0.05。結(jié)果:1.兩組患者眶脂肪中PPAR-γmRNA的表達(dá)水平:TAO組2.640±0.525,CON組1.182±0.272,TAO組明顯高于CON組,差異有統(tǒng)計(jì)學(xué)意義(P0.05)。2.眶脂肪中兩組患者PPAR-γ蛋白的表達(dá)水平:TAO組0.992±0.104,CON組0.634±0.0291,T AO組明顯高于CON組,差異有統(tǒng)計(jì)學(xué)意義(P0.05)。3.血清中四組患者PPAR-γ的表達(dá)水平:活動(dòng)組0.0133±0.0031pg/ml,非活動(dòng)組0.00903±0.00421bpg/ml,GD組0.0190±0.00323pg/ml,CON組0.00813±0.00774 pg/ml。四組比較,差異有統(tǒng)計(jì)學(xué)意義(F=16.199,P0.001),其中活動(dòng)組與非活動(dòng)組比較,活動(dòng)組明顯高于非活動(dòng)組,差異有統(tǒng)計(jì)學(xué)意義(P=0.0150.05);活動(dòng)組與GD組比較,活動(dòng)組明顯低于GD組,差異有統(tǒng)計(jì)學(xué)意義(P=0.001);活動(dòng)組與CON組比較,活動(dòng)組明顯高于CON組,差異有統(tǒng)計(jì)學(xué)意義(P=0.0030.05);非活動(dòng)組與GD組比較,非活動(dòng)組明顯低于G D組,差異有統(tǒng)計(jì)學(xué)意義(P0.001);非活動(dòng)組與CON組比較,差異無統(tǒng)計(jì)學(xué)意義(P=0.6100.05);GD組與CON組比較,GD組明顯高于CON組,差異有統(tǒng)計(jì)學(xué)意義(P0.001)。男女患者血清中PPAR-γ表達(dá)水平無統(tǒng)計(jì)學(xué)差異(P0.05),不同年齡患者血清中PPAR-γ表達(dá)水平亦無統(tǒng)計(jì)學(xué)差異(P0.05)。結(jié)論:1.TAO患者眶脂肪PPAR-γ基因轉(zhuǎn)錄水平及蛋白表達(dá)水平均顯著增加。2.活動(dòng)期TAO患者血清中PPAR-γ表達(dá)水平顯著增高。3.甲狀腺功能亢進(jìn)患者血清中PPAR-γ表達(dá)水平顯著增高。4.年齡和性別對(duì)TAO患者血清中PPAR-γ的表達(dá)無顯著影響。
[Abstract]:Objective: to detect the expression of PPAR- 緯 in orbital adipose tissue and serum of TAO patients, and to explore the role of PPAR- 緯 in the pathogenesis of TAO. The expression of PPAR- 緯 mRNA in orbital fat and the expression of PPAR- 緯 protein in orbital fat were detected by RT-PCR method and Western blot method respectively. 19 female patients were divided into two groups according to the clinical activity score (CAS) and 99Tcm-DTPA orbital imaging: 18 cases (male 7 cases, female 11 cases) with CAS399Tcm-DTPA orbital imaging positive and 15 cases (male 7 cases) with CAS399Tcm-DTPA negative orbital imaging (male 7 cases), with CAS399Tcm-DTPA imaging negative (7 cases), the patients with CAS399Tcm-DTPA orbital imaging were divided into two groups: CAS399Tcm-DTPA imaging was positive in 18 cases (male 7 cases, female 11 cases). There were 16 patients (6 males, 10 females) with Graves disease and 15 healthy controls (7 males and 8 females). The expression of PPAR- 緯 in serum of the four groups was detected by Elisa method. SPSS19.0 was used for statistical analysis, and the measured data were expressed as x 鹵s. The counting data are expressed by examples (composition ratio), the mean of normal distribution data is compared by ANOVA, the non-normal distribution data mean is compared by rank sum test, and the composition ratio is compared by chi-square test. Results the expression level of PPAR- 緯 mRNA in the orbital fat of the two groups was significantly higher than that in the CON group (2.640 鹵0.525) and 1.182 鹵0.272 respectively. The expression level of PPAR- 緯 protein in the two groups was significantly higher than that in the CON group (0.634 鹵0.0291T AO, 0.992 鹵0.104CON, 0.634 鹵0.0291T). The expression level of PPAR- 緯 in the four groups was 0.0133 鹵0.0031 PG / ml in the active group, 0.00903 鹵0.00421 bpg / ml in the inactive group, 0.0190 鹵0.00323pg / ml / ml group in the GD group, 0.00813 鹵0.00774 pg / ml / ml in the four groups. There was significant difference in the expression of PPAR- 緯 between the active group and the inactive group, and the difference between the active group and the inactive group was significantly higher than that in the inactive group, and the expression of PPAR- 緯 in the active group was significantly higher than that in the inactive group. The difference between active group and GD group was significantly lower than that in GD group, the difference was statistically significant, and the difference between active group and CON group was significantly higher than that in CON group (P 0.0030.05), the difference between active group and GD group was statistically significant, and the difference between inactive group and GD group was significantly higher than that in active group and GD group. There was no significant difference between the inactive group and the CON group, and the difference between the GD group and the CON group was higher than that in the CON group. There was no significant difference in the expression of PPAR- 緯 between male and female patients and there was no significant difference in the expression of PPAR- 緯 between male and female patients with different ages. Conclusion the transcription level and protein expression of PPAR- 緯 gene in orbital fat of patients with TAO are not significantly different from those of patients of different ages. The expression of PPAR- 緯 in serum of patients with hyperthyroidism was significantly higher than that of patients with hyperthyroidism. 4. Age and sex had no significant effect on the expression of PPAR- 緯 in serum of patients with TAO.
