神經(jīng)生長(zhǎng)因子在糖尿病大鼠視網(wǎng)膜突觸可塑性中的作用

【摘要】:目的探討神經(jīng)生長(zhǎng)因子(nerve growth factors,NGF)在糖尿病大鼠視網(wǎng)膜突觸可塑性中的作用。方法取清潔級(jí)雄性SD大鼠24只,隨機(jī)分為對(duì)照組、糖尿病組、治療組,每組8只。后兩組采用鏈脲佐菌素誘導(dǎo)糖尿病模型,模型誘導(dǎo)成功后,治療組給予腹腔注射鼠NGF(800 U·kg-1),每天1次。對(duì)照組、糖尿病組給予等劑量生理鹽水。12周后,檢測(cè)丙二醛(MDA)含量及超氧化物歧化酶(SOD)活性,免疫熒光技術(shù)檢測(cè)視網(wǎng)膜突觸素表達(dá),Western blot檢測(cè)視網(wǎng)膜突觸素、Caspase-3的表達(dá)。結(jié)果3組間MDA含量比較,總體差異有統(tǒng)計(jì)學(xué)意義(F=85.46,P0.01);糖尿病組MDA含量較對(duì)照組明顯升高(P0.01),治療組較糖尿病組明顯降低(P0.01)。3組間SOD活性比較,總體差異有統(tǒng)計(jì)學(xué)意義(F=17.76,P0.01);糖尿病組SOD活性較對(duì)照組明顯降低(P0.01),治療組較糖尿病組明顯升高(P0.01)。3組間突觸素免疫熒光強(qiáng)度比較,總體差異有統(tǒng)計(jì)學(xué)意義(F=395.42,P0.01),糖尿病組較對(duì)照組熒光強(qiáng)度明顯下降(P0.01),治療組較糖尿病組熒光強(qiáng)度明顯升高(P0.01)。3組間突觸素相對(duì)表達(dá)量比較,總體差異有統(tǒng)計(jì)學(xué)意義(F=17.27,P0.01)。糖尿病組較對(duì)照組突觸素表達(dá)明顯減少(P0.01),治療組較糖尿病組明顯增多(P0.01)。Caspase-3蛋白相對(duì)表達(dá)量3組間比較,總體差異有統(tǒng)計(jì)學(xué)意義(F=217.13,P0.01)。糖尿病組Caspase-3表達(dá)較對(duì)照組明顯增多(P0.01),治療組較糖尿病組明顯減少(P0.01)。結(jié)論NGF通過(guò)降低糖尿病視網(wǎng)膜氧化應(yīng)激,抑制了細(xì)胞凋亡,恢復(fù)了視網(wǎng)膜突觸數(shù)量。提示NGF可能通過(guò)氧化應(yīng)激途徑參與了糖尿病視網(wǎng)膜突觸可塑性的變化。
[Abstract]:Objective to investigate nerve growth factor growth factors. Methods Twenty-four male SD rats of clean grade were randomly divided into control group, diabetic group and treatment group. The diabetic model was induced by streptozotocin (STZ) in the last two groups. After the model was successfully induced, the treatment group was given intraperitoneal injection of NGF(800 U 路kg -1, once a day, in the control group. The content of malondialdehyde (MDA) and the activity of superoxide dismutase (SOD) were measured in diabetic group after 12 weeks of administration of equal dose of normal saline. The expression of synaptophysin in retina was detected by immunofluorescence technique. Western blot was used to detect the expression of Caspase-3 in retina synaptophysin. Results the MDA content of the three groups was significantly higher than that of the control group (85.46). P0.01; Compared with the control group, the MDA content in the diabetic group was significantly higher than that in the control group, and the activity of SOD in the treatment group was significantly lower than that in the diabetic group. The overall difference was statistically significant (P 0.01). Compared with the control group, the activity of SOD in the diabetic group was significantly lower than that in the control group, and the immunofluorescence intensity of synaptophysin in the treatment group was significantly higher than that in the diabetic group. The total difference was statistically significant (P 0.01). The fluorescence intensity of diabetic group was significantly lower than that of control group (P 0.01). Compared with the diabetic group, the relative expression of synaptophysin in the treatment group was significantly higher than that in the diabetic group. The overall difference was statistically significant. The expression of synaptophysin in diabetic group was significantly lower than that in control group (P 0.01). Compared with the diabetic group, the relative expression of P0.01U. Caspase-3 protein in the treatment group was significantly higher than that in the diabetic group. The total difference was statistically significant among the three groups. The expression of Caspase-3 in diabetic group was significantly higher than that in control group (P 0.01). Conclusion NGF can inhibit apoptosis by reducing oxidative stress of diabetic retina. It suggests that NGF may be involved in the changes of synaptic plasticity in diabetic retina through oxidative stress pathway.
