【摘要】：目的:探討聯(lián)合抑制缺氧誘導(dǎo)因子1α(HIF-1α)和信號(hào)轉(zhuǎn)導(dǎo)子和轉(zhuǎn)錄激活子3(STAT3)對(duì)裸鼠人喉鱗狀細(xì)胞癌移植瘤化療增敏的作用。方法:將制備的喉癌移植瘤裸鼠隨機(jī)分為空白對(duì)照組(A組)、順鉑化療組(B組)、順鉑化療聯(lián)合AG490組(C組)、順鉑化療聯(lián)合HIF-1α~(-/-)組(D組)、順鉑化療聯(lián)合AG490加HIF-1α~(-/-)組(E組)和HIF-1α~(-/-)組(F組)。給予腹腔注射順鉑3 mg/kg連續(xù)3d,然后間隔給予順鉑和10mg/kg AG490連續(xù)12d。應(yīng)用免疫組織化學(xué)法檢測(cè)Ki67和HIF-1α表達(dá)。應(yīng)用Western blot法檢測(cè)p-STAT3表達(dá)。結(jié)果:C組、D組與B組比較HIF-1α表達(dá)均有顯著差異(t值分別為2.782、3.873,均P0.05);E組與C組、D組比較HIF-1α表達(dá)均有顯著差異(t值分別為6.140、3.667,均P≤0.01)。C組與B組比較p-STAT3表達(dá)明顯降低(t=17.840,P0.01);D組與B組比較p-STAT3表達(dá)無(wú)明顯差異(t=0.038,P0.05);E組與C組、D組比較p-STAT3表達(dá)明顯降低(均P0.01)。E組與B組比較抑瘤率有顯著差異(t=5.509,P0.01);E組與C組、D組比較抑瘤率均有顯著差異(t值分別為3.422、2.718,均P0.05)。E組與B組、D組比較Ki67指數(shù)有顯著差異(t值分別為8.307、4.524,均P0.01);E組與C組比較Ki67指數(shù)有顯著差異(t=3.736,P0.05)。結(jié)論:聯(lián)合抑制HIF-1α表達(dá)和STAT3通路對(duì)于頭頸鱗狀細(xì)胞癌的增殖抑制作用明顯較單獨(dú)抑制一種通路強(qiáng)。
[Abstract]:Objective: to investigate the combined inhibition of hypoxia inducible factor-1 偽 (HIF-1 偽) and signal transducers and activators of transcription (3STAT3). Methods: the nude mice were randomly divided into control group (group A). Group B was treated with cisplatin, group C with cisplatin combined with AG490, group D with cisplatin combined with HIF-1 偽 -r-r). Cisplatin chemotherapy combined with AG490 plus HIF-1 偽 -r-r-group (group E) and HIF-1 group (group F) were given intraperitoneal injection of cisplatin for 3 mg/kg for 3 days. Cisplatin and 10mg / kg AG490 were given at intervals for 12 days. The expression of Ki67 and HIF-1 偽 was detected by immunohistochemical method. Western was used. The expression of p-STAT3 was detected by blot. The expression of HIF-1 偽 in group D was significantly different from that in group B (2.782 鹵3.873, P 0.05). There was significant difference in the expression of HIF-1 偽 between group E and group C (6.140 鹵3.667). The expression of p-STAT3 in group C was significantly lower than that in group B (P 鈮，
