川芎嗪抑制人視網(wǎng)膜母細(xì)胞瘤WERI-Rb1細(xì)胞增殖的機制研究

【摘要】:目的探索川芎嗪(TMP)對WERI-Rb1細(xì)胞增殖的影響及TMP下調(diào)人WERI-Rb1細(xì)胞中趨化因子受體4(CXCR4)的轉(zhuǎn)錄的分子機制。方法 (1)TMP及CXCR4的拮抗劑AMD3100藥物分別處理WERIRb1細(xì)胞,相同濃度PBS及DMSO作為對照,活細(xì)胞成像儀觀察WERI-Rb1細(xì)胞形態(tài)及密度變化。(2)WERIRb1細(xì)胞經(jīng)TMP處理后,采用熒光定量逆轉(zhuǎn)錄PCR、免疫熒光及蛋白免疫印跡法分別從mRNA和蛋白質(zhì)水平檢測TMP對WERI-Rb1細(xì)胞CXCR4及NRF1表達(dá)的影響。結(jié)果 (1)活細(xì)胞成像儀觀察發(fā)現(xiàn)TMP處理組與AMD3100處理組的細(xì)胞狀態(tài)較正常組差,增殖明顯受抑制。(2)熒光定量逆轉(zhuǎn)錄PCR結(jié)果顯示:與對照組比較,TMP處理組WERI-Rb1細(xì)胞中CXCR4和NRF1mRNA表達(dá)均明顯下降(P0.05);免疫熒光及蛋白免疫印跡實驗結(jié)果顯示,與對照組比較,TMP處理組WERI-Rb1細(xì)胞中CXCR4和NRF1蛋白表達(dá)均明顯下降(P0.05)。結(jié)論TMP對WERI-Rb1細(xì)胞的增殖有明顯抑制作用,且能下調(diào)WERI-Rb1細(xì)胞中CXCR4及NRF1的表達(dá)水平。
[Abstract]:Objective to explore the effect of tetramethylpyrazine (TMP) on the proliferation of WERI-Rb1 cells and WERI-Rb1 cells in TMP down-regulation of chemokine receptor 4 (CXCR4) of the molecular mechanism of transcription. Methods (1) TMP and CXCR4 antagonist AMD3100 drugs were treated WERIRb1 cells, the same concentration of PBS and DMSO as control, to observe the changes of live cell imaging instrument the shape and density of WERI-Rb1 cells. (2) WERIRb1 cells after treatment with TMP by fluorescence quantitative reverse transcription PCR, immune fluorescence and protein immunoblotting method respectively from the impact of the level of mRNA and protein of WERI-Rb1 cells to detect the expression of TMP CXCR4 and NRF1. Results (1) live cell imaging instrument observation TMP treatment group and AMD3100 treatment group. The status of cells compared with the normal group, the proliferation was obviously inhibited. (2) fluorescence quantitative reverse transcriptase PCR results showed that: compared with the control group, TMP treatment group WERI-Rb1 cells CXCR4 and NRF1mRNA expression were significantly decreased (P0.05); The results of immunofluorescence and Western blot showed that, compared with the control group, TMP treatment group WERI-Rb1 cells CXCR4 and NRF1 protein expression were significantly decreased (P0.05). Conclusion TMP can significantly inhibit the proliferation of WERI-Rb1 cells, the expression level of NRF1 and down-regulation of CXCR4 and WERI-Rb1 cells.
