低氧預(yù)處理和七氟醚預(yù)處理對成年大鼠短暫性全腦缺血的保護(hù)作用及對Jun-B mRNA表達(dá)的影響

目的:觀察低氧預(yù)處理和七氟醚預(yù)處理對成年大鼠短暫性全腦缺血(Transient global cerebral ischemia,tGCI)的保護(hù)作用及對Jun-B mRNA表達(dá)的影響。方法:成年健康雄性(Sprague Dawley,SD)大鼠,48只,體重240~250g,隨機(jī)分4組:全腦缺血再灌注組(tGCI組),低氧預(yù)處理(Hypoxic preconditioning,HPC)組,七氟醚預(yù)處理(Sevoflurane,SEV)組,低氧+七氟醚預(yù)處理組(H+S組),每組大鼠12只。tGCI組:用Pulsinelli四血管閉塞法(4VO)建立10 min tGCI大鼠模型;HPC組:反復(fù)暴露于減壓艙內(nèi)模擬海拔4000m,每天1 h,連續(xù)5 d;SEV組:大鼠接受2.4%七氟醚與100%氧氣混合氣體的預(yù)處理,1h/d,連續(xù)5d;H+S組:低氧預(yù)處理聯(lián)合七氟醚預(yù)處理,連續(xù)5天,1 h/d。缺血再灌注4 h(T1)、24 h(T2)、7 d(T3)分別處死大鼠,制作海馬組織冰凍切片,行(Fluoro-Jade B,FJB)熒光染色,觀察海馬組織神經(jīng)細(xì)胞凋亡,通過Trizol提取海馬組織...
