CRLF1激活ERK1/2信號(hào)通路調(diào)控TGF-β1介導(dǎo)的黃韌帶肥厚的機(jī)制研究

背景黃韌帶肥厚是腰椎椎管狹窄的一個(gè)重要因素,嚴(yán)重時(shí)會(huì)壓迫神經(jīng)根和馬尾,導(dǎo)致腰部疼痛及間歇性跛行。盡管有研究認(rèn)為黃韌帶肥厚可能和各種炎癥因子、生長(zhǎng)因子、老齡及機(jī)械應(yīng)力有關(guān),但是導(dǎo)致黃韌帶肥厚的關(guān)鍵分子及其調(diào)控機(jī)制仍然未知。另外,由于缺乏有效的黃韌帶肥厚動(dòng)物模型,尚未發(fā)現(xiàn)關(guān)于黃韌帶肥厚分子機(jī)制的體內(nèi)研究。目的本研究旨在:1.發(fā)現(xiàn)黃韌帶肥厚的關(guān)鍵調(diào)控蛋白,探索其調(diào)控細(xì)胞外基質(zhì)形成的機(jī)制。2.建立小鼠黃韌帶肥厚模型,對(duì)黃韌帶肥厚的分子機(jī)制進(jìn)行體內(nèi)驗(yàn)證。方法1.收集正常和肥厚的人黃韌帶組織,組織學(xué)染色檢測(cè)兩組纖維化情況。用同位素相對(duì)標(biāo)記與絕對(duì)定量(isobaric tags for relative and absolute quantitation,iTRAQ)技術(shù)對(duì)兩組樣本進(jìn)行分析,整合轉(zhuǎn)錄組與蛋白質(zhì)組數(shù)據(jù),發(fā)現(xiàn)關(guān)鍵蛋白:細(xì)胞因子受體樣因子1(Cytokine Receptor Like Factor 1,CRLF1)。使用免疫組化和qPCR技術(shù)對(duì)結(jié)果進(jìn)行驗(yàn)證。2.分離并體外培養(yǎng)人正常黃韌帶細(xì)胞,使用TGF-β1和成熟的重組人CRLF1刺激黃韌帶細(xì)胞,通過(guò)免疫熒光、Westen Blot及q...
