新芒果苷防治破骨細(xì)胞相關(guān)性骨溶解疾病的機(jī)制研究

骨的穩(wěn)態(tài)是骨吸收與骨形成的一個(gè)動(dòng)態(tài)平衡,而這一穩(wěn)態(tài)主要由成骨細(xì)胞的成骨作用和破骨細(xì)胞的骨吸收作用來(lái)維持。成骨細(xì)胞作為骨形成的主要功能細(xì)胞,主要來(lái)源于骨膜和骨髓基質(zhì)的間充質(zhì)始祖細(xì)胞。分化成熟的成骨細(xì)胞位于骨的表面,產(chǎn)生粘多糖、糖蛋白膠原纖維和膠原等,促進(jìn)細(xì)胞外骨基質(zhì)礦化形成骨細(xì)胞。在骨重塑過(guò)程中,破骨細(xì)胞和成骨細(xì)胞間的相互作用發(fā)生在基本多細(xì)胞單位(basic multicellular unit),通過(guò)直接接觸、分泌旁分泌因子及細(xì)胞與骨基質(zhì)作用等方式進(jìn)行相互調(diào)節(jié),進(jìn)而精確調(diào)控骨的重塑過(guò)程。在成骨細(xì)胞主要表現(xiàn)為分泌大量細(xì)胞因子(如Ⅰ型膠原、骨鈣素等),刺激破骨細(xì)胞的形成分化及促進(jìn)功能的改變,對(duì)破骨前體細(xì)胞的遷徙進(jìn)行調(diào)節(jié)干預(yù)。破骨細(xì)胞是由骨髓中的髓系祖細(xì)胞分化而成的單核巨噬細(xì)胞相互融合形成,是一種多核巨細(xì)胞。成熟的破骨細(xì)胞主要行使骨的溶解吸收功能,同時(shí)亦有調(diào)控骨內(nèi)血管形成、調(diào)控骨形成和骨鈣素的激素作用等生物學(xué)作用。破骨細(xì)胞受骨保護(hù)素、RANK配體、甲狀旁腺激素、降鈣素、白細(xì)胞介素6等多種因子的影響和調(diào)節(jié)。破骨細(xì)胞形成分化需要核因子κB配體的受體活化因子配體(RANKL)和巨噬細(xì)胞集落刺激因子...
