不同劑量姜黃素腹腔注射對(duì)脊髓損傷大鼠脊髓組織結(jié)構(gòu)及后肢運(yùn)動(dòng)功能的改善作用
[Abstract]:Objective to observe the effects of different doses of curcumin on spinal cord tissue structure and motor function in (SCI) rats with spinal cord injury. Methods 35 SD rats were randomly divided into curcumin high dose group, curcumin medium dose group, curcumin low dose group, SCI group and control group. The SCI model was established by HI-0400 spinal cord percussion apparatus in 4 groups except the control group. Rats in high, middle and low dose curcumin groups were intraperitoneally injected with 200, 100, 50 mg/kg curcumin 30 min after SCI. The spinal cord tissue of each group was observed by HE staining on the 7th day after sci. BBB score and modified Rivlin oblique plate test were performed to evaluate the motor function of the hindlimb of the rats in each group on the 7th day after sci. On the 7th, 14th, 21st and 28th day, the motor function of the hindlimb of the rats in each group was evaluated by routine feeding and intraabdominal injection of the same amount of saline at the same time point. Results focal hemorrhage of spinal cord was observed 1 day after injury in SCI group, the boundary between gray matter and white matter was not clear, and many neurons converged, there were more vacuoles in cytoplasm, and the infiltration of inflammatory cells was accompanied by the infiltration of inflammatory cells. On the 3rd day after injury, the amount of bleeding in the injured area decreased, fat liquefaction, nuclear fragmentation, a large number of cells dissolved and disappeared, most of the nucleus atrophy disappeared, the gray matter center obviously died, the axonal disorder in the gray matter was irregular, more vacuoles were formed in the cytoplasm, and the apoptosis was obviously increased. On the 7th day after injury, the degree of SCI was improved and the bleeding phenomenon disappeared basically. Although a small amount of liquefaction necrosis, vacuole and cell loss were found in the gray matter and white matter, the loss of cells was alleviated. Compared with the SCI group, the morphology of spinal cord tissue in the high, middle and low dose groups was improved, the degeneration and necrosis of spinal cord neurons and the infiltration of inflammatory cells in spinal cord tissue were less, and the number of neuronal apoptosis was less. 7 days after sci, 14, 21, 28 days after sci, the BBB score and the maximum oblique plate angle of Rivlin oblique plate test in sci group were significantly lower than those in control group (P 0.05). With the passage of time, the BBB score and the maximum oblique plate angle of SCI group and curcumin group increased gradually after SCI, and the maximum oblique plate angle of Rivlin oblique plate test increased gradually. At the same time, the BBB score and the maximum oblique plate angle of curcumin in the middle dose group were lower than those in the control group (P < 0.05), and the curcumin in the high dose group was lower than that in the curcumin middle dose group (P < 0.05), SCI), and that in the high dose group was lower than that in the high dose group (P < 0.05). Conclusion Intraabdominal injection of curcumin can promote the recovery of spinal cord tissue structure and motor function in SCI rats, and 100 g/kg curcumin has the best effect on the recovery of spinal cord motor function in SCI rats.
