脂肪干細(xì)胞覆膜聚丙烯補(bǔ)片在大鼠體內(nèi)的生物相容性

【摘要】:目的探討脂肪干細(xì)胞(ADSCs)覆膜聚丙烯補(bǔ)片在大鼠體內(nèi)的生物相容性。方法培養(yǎng)擴(kuò)增檢測(cè)大鼠脂肪干細(xì)胞后,將其接種到聚丙烯補(bǔ)片上,體外培養(yǎng)10 d。成年雄性SD大鼠12只,個(gè)體質(zhì)量約(250±20)g,隨機(jī)分為覆膜補(bǔ)片組和未覆膜補(bǔ)片組,每組6只。分別將聚丙烯補(bǔ)片和ADSCs覆膜聚丙烯補(bǔ)片植入大鼠的腹壁肌肉表面,30 d后觀察補(bǔ)片侵蝕、粘連情況,測(cè)量補(bǔ)片厚度、硬度、韌性等,并進(jìn)行組織學(xué)觀察。結(jié)果細(xì)胞呈長(zhǎng)梭形,并形成小集落,呈束狀、菊花狀或旋渦狀生長(zhǎng),活力良好,免疫熒光檢測(cè)結(jié)果顯示干細(xì)胞特異性標(biāo)記CD90表達(dá)≥97%。ADSCs在聚丙烯補(bǔ)片上附著生長(zhǎng)良好。植入大鼠體內(nèi)30 d后,ADSCs覆膜聚丙烯補(bǔ)片炎癥反應(yīng)較聚丙烯補(bǔ)片輕,炎癥細(xì)胞評(píng)分[(0.95±0.23)分vs(2.62±0.40)分,P0.05],而成纖維細(xì)胞增殖和新生血管生成卻更多,分別為[(2.6±0.4)分vs(1.8±0.4)分,P0.05]和[(2.4±0.4)分vs(1.5±0.4)分,P0.05]。30 d后ADSCs覆膜聚丙烯補(bǔ)片的硬度與聚丙烯補(bǔ)片無(wú)明顯差別[(2.1±0.3)分vs(2.2±0.3)分,P0.05],而厚度[(2.67±0.37)分vs(1.13±0.24)分,P0.05]、韌性[(2.67±0.31)分vs(2.27±0.26)分,P0.05]等均大于聚丙烯補(bǔ)片。結(jié)論 ADSCs覆膜聚丙烯補(bǔ)片相較聚丙烯補(bǔ)片植入大鼠體內(nèi)后可以減輕局部炎癥反應(yīng)、增加局部腹壁順應(yīng)性。
[Abstract]:Objective to investigate the biocompatibility of adipose stem cell (ADSCs) coated polypropylene patch in rats. Methods after the rat adipose stem cells were detected by culture amplification, the adipose stem cells were inoculated on polypropylene patch and cultured in vitro for 10 days. Twelve adult male SD rats, with an individual mass of about (250 鹵20) g, were randomly divided into two groups: covered patch group (n = 6) and uncoated patch group (n = 6). Polypropylene patch and ADSCs coated polypropylene patch were implanted into the muscle surface of abdominal wall of rats, 30 days later, the erosion and adhesion of patch were observed, the thickness, hardness and toughness of patch were measured, and the histology was observed. Results the cells were long fusiform and formed small colonies, bundles, chrysanthemum or vortex growth, and good vitality.
