蛛網(wǎng)膜下腔注射布比卡因?qū)Υ笫蠹顾鐶ABA-AR alpha1亞基的影響

【摘要】:目的 觀察蛛網(wǎng)膜下腔注射布比卡因?qū)Υ笫蠹顾柩虼螃?氨基丁酸A受體alphal(GABA-ARα1)亞基表達(dá)的影響,探討布比卡因椎管內(nèi)注射產(chǎn)生鎮(zhèn)靜作用的機(jī)制。方法雄性SD大鼠32只,體重260±20g,隨機(jī)分為4組:空白對(duì)照組(C組),鹽水對(duì)照組(NC組),0.5%布比卡因10min組(B10組),0.5%布比卡因30min組(B30組),每組8只。C組不做任何處理,取材同下組;其余三組建立大鼠蛛網(wǎng)膜下腔置管模型后,注射生理鹽水或0.5%布比卡因20ul,于相應(yīng)時(shí)點(diǎn)斷頸椎處死取脊髓腰膨大段,分別進(jìn)行尼氏染色、免疫熒光染色,觀察脊髓神經(jīng)細(xì)胞形態(tài)、GABA-ARα1蛋白在脊髓的分布及檢測(cè)脊髓Ⅱ板層平均光密度的變化;用western-blot檢測(cè)GABA-ARα1亞基蛋白在總蛋白、細(xì)胞漿蛋白及細(xì)胞膜蛋白中表達(dá)的變化。結(jié)果 1.尼氏染色結(jié)果,鏡下可見(jiàn)各組神經(jīng)元細(xì)胞排列整齊,未見(jiàn)神經(jīng)元形態(tài)改變;2.免疫熒光結(jié)果顯示,GABA-ARα1蛋白主要分布于脊髓灰質(zhì)各板層,高倍鏡下可見(jiàn)細(xì)胞漿及細(xì)胞膜上均有該蛋白存在,以Ⅱ板層分布最為密集,各組之間脊髓Ⅱ板層GABA-ARα1平均光密度值無(wú)顯著改變(P0.05);3.western-blot檢測(cè)結(jié)果顯示,各組GABA-ARα1亞基在總蛋白中相對(duì)表達(dá)量無(wú)顯著差異(P0.05);各組細(xì)胞漿蛋白中GABA-ARα1亞基表達(dá)量:B10組、B30組與C組及NC組相比均顯著下降(p0.01),且B10組下降更明顯;各組細(xì)胞膜蛋白中GABAARα1亞基表達(dá)量:B10組、B30組、與C組及NC組相比均顯著上調(diào)(p0.01),且B10組上調(diào)更明顯;結(jié)論 1.蛛網(wǎng)膜下腔注射0.5%布比卡因使GABA-ARα1亞基蛋白從細(xì)胞漿轉(zhuǎn)運(yùn)至細(xì)胞膜,上調(diào)細(xì)胞膜該受體的表達(dá)數(shù)量,這可能是椎管阻滯引起鎮(zhèn)靜作用的機(jī)制之一。2.蛛網(wǎng)膜下腔注射0.5%布比卡因在作用10min時(shí)對(duì)GABA-ARα1亞基蛋白細(xì)胞膜表達(dá)的影響最大。
[Abstract]:Aim to observe the effect of bupivacaine injection into subarachnoid space on the expression of 緯-aminobutyric acid A receptor alphal (GABA-AR 偽 1) subunit in spinal cord of rats, and to explore the mechanism of sedative effect induced by bupivacaine intraspinal injection. Methods 32 male SD rats, weighing 260 鹵20g, were randomly divided into 4 groups: blank control group (C group), saline control group (NC group), 0.5% bupivacaine 10min group (B10 group) and 0.5% bupivacaine 30min group (B30 group). There were 8 rats in each group. Group C did not do any treatment, and the material was taken from the same group as the lower group. The other three groups were injected with normal saline or 0.5% bupivacaine 20ul.After establishing the subarachnoid catheterization model in the other three groups, the spinal cord lumbar enlargement segments were killed at the corresponding time point and stained by Nissl's staining and immunofluorescence staining, respectively.
