坐骨神經(jīng)成纖維細胞修復周圍神經(jīng)缺損

【摘要】:坐骨神經(jīng)成纖維細胞是正常神經(jīng)組織中含量豐富的細胞,有著重要的功能作用。有研究表明乳腺等組織中間質成纖維細胞和其上皮共移植可以更好的促進組織器官的重塑。周圍神經(jīng)組織中的成纖維細胞對于周圍神經(jīng)再生起著何種作用,目前尚不明確。目的:我們想探究在周圍神經(jīng)離斷傷后,其組織中的成纖維細胞在神經(jīng)再生過程中的作用及其可能的機制。方法:本研究擬通過單純的坐骨神經(jīng)成纖維細胞移植或者坐骨神經(jīng)成纖維細胞和許旺細胞共移植能否更好的促進神經(jīng)的再生。首先,用PUR導管構建一個5mm神經(jīng)缺損模型,將六組細胞分別注入導管并回植橋接坐骨神經(jīng)缺損;六組細胞分別是:純matrigel組(control blank),坐骨神經(jīng)成纖維細胞組(SN-Fb),皮膚成纖維細胞組(Dm-Fb)坐骨神經(jīng)成纖維細胞和許旺細胞1:1混合組(SN-FbSC1:1),坐骨神經(jīng)成纖維細胞和許旺細胞1:2混合組(SN-FbSC1:2),坐骨神經(jīng)許旺細胞組(SN-SC)。三個月后,對再生的神經(jīng)進行組織學檢測(軸突、許旺細胞及細胞外基質的免疫熒光染色)和靶器官功能學的檢測(肌肉濕重統(tǒng)計及Masson染色)。結果:單純的坐骨神經(jīng)成纖維細胞回植產(chǎn)生過多的細胞外基質占位,阻礙神經(jīng)再生。Fb與SC以1:2比例回植到缺損的神經(jīng)中,從再生的神經(jīng)軸突統(tǒng)計和腓腸肌萎縮及纖維化程度上,均和單純的SC回植無差異,在再生許旺細胞數(shù)量的統(tǒng)計中優(yōu)于單純的SC回植。在體外細胞分泌的神經(jīng)營養(yǎng)因子檢測中,SN-FbSC1:2組均高于SN-SC組。結論:Fb與SC以1:2比例回植到缺損的神經(jīng)中,修復效果優(yōu)于單純的許旺細胞回植,因此Fb在神經(jīng)損傷修復過程中可能與SC起到協(xié)同作用,共同促進神經(jīng)軸突的再生。
[Abstract]:Sciatic nerve fibroblasts are abundant cells in normal nerve tissue and have important function. Studies have shown that cotransplantation of stromal fibroblasts and their epithelium can promote the remodeling of tissues and organs. The role of fibroblasts in peripheral nerve regeneration is unclear. Aim: to explore the role of fibroblasts in the process of nerve regeneration and its possible mechanism after peripheral nerve injury. Methods: in this study, whether the sciatic nerve fibroblast transplantation or the sciatic nerve fibroblast and Schwann cell co-transplantation can better promote nerve regeneration. Firstly, a model of 5mm nerve defect was constructed with PUR catheter. The six groups of cells were injected into the catheter and transplanted back to graft the sciatic nerve defect. The six groups of cells were: (control blank), sciatic nerve fibroblast group (SN-Fb), skin fibroblast group (Dm-Fb), sciatic nerve fibroblast and Schwann cell 1:1 mixture group (SN-FbSC1:1). Sciatic nerve fibroblasts and Schwann cells were mixed at 1:2 (SN-FbSC1:2) and sciatic nerve Schwann cells (SN-SC). Three months later, the regenerated nerves were examined for histology (immunofluorescence staining of axons, Schwann cells and extracellular matrix) and for the function of target organs (muscle wet weight statistics and Masson staining). Results: the reimplantation of sciatic nerve fibroblasts resulted in too much extracellular matrix occupation, which blocked the regeneration of nerve. Fb and SC were implanted back into the defective nerve at 1:2. From the statistics of regenerated nerve axons and the degree of gastrocnemius atrophy and fibrosis, there was no difference between the regenerated nerve axons and the degree of atrophy and fibrosis of gastrocnemius muscle, which was superior to that of SC replantation in the statistics of the number of regenerated Schwann cells. The level of neurotrophic factor in SN-FbSC1:2 group was higher than that in SN-SC group. Conclusion: the repair effect of Fb and SC is better than that of Schwann cell replantation in proportion of 1:2 to the defective nerve. So Fb may play a synergistic role with SC in the repair of nerve injury and promote the regeneration of nerve axons.
