右美托咪定對(duì)糖尿病大鼠體液免疫及補(bǔ)體的影響

【摘要】:目的:通過(guò)腹腔注射鏈脲佐菌素建立糖尿病大鼠模型,觀察右美托咪定對(duì)糖尿病大鼠體液免疫功能及補(bǔ)體功能的影響,探討右美托咪定在糖尿病大鼠體液免疫功能調(diào)節(jié)中的作用。方法:選擇健康成年雄性SD大鼠72只,10~12周齡,體重300~350g,采用隨機(jī)數(shù)字表法,將其隨機(jī)均分為3組:對(duì)照組(C組)、糖尿病組(DM組)和右美托咪定+糖尿病組(DEX組),每組24只。DM組和DEX組采用鏈脲佐菌素(STZ)一次性腹腔注射60mg/kg建立糖尿病大鼠模型,于糖尿病模型穩(wěn)定后4W時(shí)三組大鼠均實(shí)施剖腹探查術(shù),維持手術(shù)探查時(shí)間為30min。DEX組于手術(shù)開(kāi)始前經(jīng)10min靜脈輸注右美托咪定負(fù)荷劑量1μg/kg,隨后以5μg/kg/h(容量1ml/h)的速率維持4h,C組和DM組均給予等容量生理鹽水。分別于術(shù)前(T0)、術(shù)畢、術(shù)后24h和72h時(shí)(T1~T3)每組隨機(jī)取6只大鼠采集血樣,測(cè)定血糖,并采用Elisa法檢測(cè)血清IgG、IgA及IgM和補(bǔ)體CH50、C3及C4的濃度;分別于T0~T3時(shí)采血結(jié)束后處死大鼠,處死前稱重,解剖分離脾臟組織稱重,計(jì)算脾臟指數(shù)[脾臟重量(mg)/大鼠重量(g)]。結(jié)果:(1)與C組比較,DM組和DEX組T0~T3時(shí)血糖濃度升高(P0.05);與DM組比較,DEX組T1~T3時(shí)血糖濃度下降(P0.05),T0時(shí)差異無(wú)統(tǒng)計(jì)學(xué)意義(P0.05)。(2)與C組比較,DM組和DEX組T0~T3時(shí)血清IgG、IgM濃度降低,IgA濃度升高(P0.05);與DM組比較,DEX組T1~T3時(shí)血清IgG、IgM濃度升高,IgA濃度降低(P0.05)。(3)與C組比較,DM組和DEX組T0~T3時(shí)血清CH50、C3及C4濃度均升高(P0.05);與DM組比較,DEX組T1~T3時(shí)血清CH50、C3及C4濃度均降低(P0.05)。(4)與C組比較,DM組和DEX組T0~T3時(shí)脾臟指數(shù)降低(P0.05);與DM組比較,DEX組T1~T3時(shí)脾臟指數(shù)升高(P0.05)。結(jié)論:糖尿病可誘發(fā)大鼠體液免疫功能下降及補(bǔ)體功能異常,手術(shù)創(chuàng)傷后體液免疫及補(bǔ)體功能進(jìn)一步下降,而右美托咪定可改善糖尿病大鼠圍術(shù)期體液免疫及補(bǔ)體功能。
[Abstract]:Objective: to observe the effect of dexmetomidine on humoral immune function and complement function of diabetic rats by intraperitoneal injection of streptozotocin. To investigate the role of dexmetidine in the regulation of humoral immune function in diabetic rats. Methods: 72 healthy adult male SD rats were randomly divided into three groups: control group (group C), diabetes mellitus group (DM group) and dexmetomidine diabetes group (DEX group). 24 rats in each group were treated with streptozotocin (STZ) intraperitoneal injection of streptozotocin (STZ) to establish diabetic model in DM group and DEX group. The exploration time of operation was maintained at 1 渭 g / kg by intravenous infusion of dexmetomidine through 10min before the operation in 30min.DEX group, and then at the rate of 5 渭 g/kg/h (volume 1ml/h) for 4 h. Group C and group DM were given the same volume of normal saline at the rate of 5 渭 g/kg/h (volume 1ml/h). Blood samples were collected from 6 rats in each group at pre-operation (T0), at the end of operation, 24h and 72h after operation (T1~T3), and the serum IgG,IgA and IgM, complement CH50,C3 and C4 were measured by Elisa method. The rats were killed at the end of T0~T3 and weighed before death. Spleen index [spleen weight (mg) / rat weight (g)] was measured by anatomical separation of spleen tissue. Results: (1) compared with group C, the concentration of blood glucose in DM group and DEX group was higher than that in group C (P0.05). Compared with DM group, the blood glucose concentration in DEX group decreased at T1~T3 (P0.05), but there was no significant difference at T0 (P0.05). (2). Compared with C group, the serum IgG,IgM concentration decreased and IgA concentration increased in DM group and DEX group (P0.05). Compared with the DM group, the serum IgG,IgM concentration increased and the IgA concentration decreased in the DEX group (P0.05). (3). Compared with the C group, the CH50,C3 and C4 levels in the DM group and DEX group increased significantly (P0.05). Compared with DM group, the serum CH50,C3 and C4 levels in DEX group were lower than those in C group (P0.05). (4). Compared with C group, the spleen index of DM group and DEX group was lower than that of C group (P0.05). Compared with DM group, the spleen index in DEX group was higher than that in T1~T3 group (P0.05). Conclusion: diabetes can induce decreased humoral immune function and abnormal complement function in rats. The humoral immunity and complement function of diabetic rats are further decreased after operation, while dexmetomidine can improve the humoral immunity and complement function of diabetic rats during perioperative period.
