骨細(xì)胞外基質(zhì)有機(jī)相網(wǎng)絡(luò)微納結(jié)構(gòu)的研究
[Abstract]:The microenvironment can regulate the daily activities of cells through various physical and chemical factors such as protein expression mitosis differentiation apoptosis and so on. It is indispensable to maintain the normal morphology and function of cells and tissues. Among the physical factors, the research on morphologic factors is in the ascendant. In the extracellular matrix of bone, there is a multilevel secondary network composed of collagen fiber, proteoglycan complex and other organic compounds, which plays an indispensable role in maintaining the physical and chemical properties of bone tissue and regulating various functions of various bone cells. This paper focuses on the morphology of the organic phase of the extracellular matrix of bone in order to enhance the understanding of the microenvironment of bone tissue from this point of view. Firstly, the structure of the network is characterized by atomic force microscope and scanning electron microscope. Secondly, the method of analyzing and extracting geometric characteristic parameters of random network is established based on image analysis algorithm. Then the computer geometric network model is constructed, and the micron scale external network model is constructed by electrostatic spinning. Finally, a nanoscale in vitro network model was constructed using hydrogel, and the effect of the degree of network crosslinking on the spread of mesenchymal stem cells was studied on the hydrogel model. It provides a basis for further understanding the role of bone organic multistage sub-network in the pathophysiological process and the rational design of bioengineering scaffolds.
