骨關(guān)節(jié)炎臨床表現(xiàn)特點(diǎn)及沉默信息調(diào)節(jié)因子1在其發(fā)病中的作用

【摘要】:背景:研究發(fā)現(xiàn)一些功能性基因的功能變化是引發(fā)骨關(guān)節(jié)炎的調(diào)控環(huán)節(jié),而年齡可能是其中的關(guān)鍵因素之一。目的:對(duì)近年來(lái)骨關(guān)節(jié)炎中的臨床現(xiàn)表現(xiàn)特點(diǎn)及沉默信息調(diào)節(jié)因子1在骨關(guān)節(jié)炎發(fā)生發(fā)展中的研究進(jìn)展做一概述。方法:計(jì)算機(jī)檢索相關(guān)期刊論文、Pub Med、Springer Link、Elsevier Science Direct數(shù)據(jù)庫(kù),檢索時(shí)間1987年1月至2016年1月,檢索詞分別為"骨關(guān)節(jié)炎;沉默信息調(diào)節(jié)因子1;軟骨"和"Osteoarthritis,Cartilage,Silent information regulation 1"。對(duì)納入符合標(biāo)準(zhǔn)的83條文獻(xiàn)進(jìn)行分析。結(jié)果與結(jié)論:研究表明,在多種信號(hào)通路中去乙;揎椪{(diào)控大量細(xì)胞因子的表達(dá)和生物學(xué)功能,繼而影響骨關(guān)節(jié)炎的發(fā)生和發(fā)展。沉默信息調(diào)節(jié)因子1具有調(diào)節(jié)機(jī)體代謝,抑制細(xì)胞凋亡,修復(fù)DNA損傷,延緩衰老,抵抗應(yīng)激反應(yīng)等重要作用,同時(shí)在表觀遺傳學(xué)修飾、基因沉默和信號(hào)轉(zhuǎn)導(dǎo)調(diào)節(jié)中發(fā)揮著重要的生物學(xué)功能,因此沉默信息調(diào)節(jié)因子1與各種年齡相關(guān)的疾病有關(guān),如骨關(guān)節(jié)炎、骨質(zhì)疏松癥、糖尿病和阿爾茨海默癥等。沉默信息調(diào)節(jié)因子1有望成為開(kāi)發(fā)治療骨關(guān)節(jié)炎藥物的重要靶點(diǎn),甚至可能在骨關(guān)節(jié)炎不成熟的階段,逆轉(zhuǎn)機(jī)械應(yīng)力導(dǎo)致的軟骨功能損傷。
[Abstract]:Background: it has been found that the functional changes of some functional genes are involved in the regulation of osteoarthritis, and age may be one of the key factors. Objective: to summarize the clinical features of osteoarthritis in recent years and the progress of silent information regulating factor 1 in the development of osteoarthritis. Methods: the full text database and Pub Med,Springer Link,Elsevier Science Direct database of Chinese periodicals were searched by computer from January 1987 to January 2016. The key words were "osteoarthritis", "silent information regulation factor 1" and "silent information regulation factor 1" respectively. Cartilage and Osteoarthritis,Cartilage,Silent information regulation 1. The 83 articles that meet the standard were analyzed. Results & conclusion: deacetylation can regulate the expression and biological function of cytokines in many signal pathways, and then affect the occurrence and development of osteoarthritis. Silencing information regulatory factor 1 plays an important role in regulating metabolism, inhibiting cell apoptosis, repairing DNA damage, delaying senescence, resisting stress response and so on. Gene silencing and signal transduction regulation play important biological functions, so silencing information regulatory factor 1 is associated with various age-related diseases, such as osteoarthritis, osteoporosis, diabetes and Alzheimer's disease. Silencing information regulatory factor 1 is expected to be an important target for the development of drugs for the treatment of osteoarthritis. It may even reverse the damage of cartilage function caused by mechanical stress in the immature stage of osteoarthritis.
