Micro-CT觀察重組人甲狀旁腺素在骨折愈合過程中的作用

【摘要】:目的使用Micro-CT,探討重組人甲狀旁腺激素(rh PTH)在2月齡雄性大鼠骨折愈合過程中的作用。方法應用2月齡雄性SD大鼠建立單側(cè)閉合性股骨骨折內(nèi)固定模型,隨機分為甲狀旁腺素治療組和對照組,術后分別皮下注射重組人甲狀旁腺素(1-34)10μg/(kg.d)或等劑量生理鹽水安慰劑(對照組),分別于術后第2、7、14、21和42天取雙側(cè)股骨標本,分別行X線檢查和Micro-CT觀察,并進行骨結(jié)構(gòu)的定量分析。結(jié)果術后第21天,甲狀旁腺素組骨折斷端兩側(cè)的骨痂之間連續(xù)性良好,骨折線已模糊不清。甲狀旁腺組較對照組,在術后第42天骨體積(bone Volume,BV)、骨體積分數(shù)(BV/TV)、骨密度(bone mineral density,BMD)和骨小梁模式因子(trabecular bone pattern factor,Tb.Pf)明顯高于對照組;而骨小梁分離度(trabecular separation,Tb.Sp)和各向異性的程度(degree of anisotropy,DA)低于對照組。結(jié)論低劑量重組人甲狀旁腺激素可提高大鼠骨折BV、BV/TV、Tb.Pf和BMD而降低Tb.Sp和DA,從而促進骨折的愈合。
[Abstract]:Objective to investigate the role of recombinant human parathyroid hormone (rh PTH) in fracture healing of 2 month old male rats by Micro-CT,. Methods two month old male SD rats were used to establish a unilateral closed femoral fracture model and were randomly divided into two groups: parathyroid hormone treatment group and control group. After operation, 10 渭 g / (kg.d) of recombinant human parathyroid hormone (1-34) or placebo (control group) were injected subcutaneously respectively. X-ray examination and Micro-CT observation were performed, and quantitative analysis of bone structure was carried out. Results on the 21st day after operation, the continuity of callus between the two sides of the fracture in parathyroid hormone group was good, and the fracture line was blurred. The bone volume (bone Volume,BV), bone volume fraction (BV/TV), bone mineral density (bone mineral density,BMD) and trabecular model factor (trabecular bone pattern factor,Tb.Pf) in parathyroid gland group were significantly higher than those in control group. The bone trabecular separation (trabecular separation,Tb.Sp) and anisotropic degree (degree of anisotropy,DA) were lower than those in the control group. Conclusion low dose recombinant human parathyroid hormone can increase BV,BV/TV,Tb.Pf and BMD, decrease Tb.Sp and DA, and promote fracture healing in rats.
