骨形態(tài)發(fā)生蛋白2和富血小板血漿凝膠對(duì)兔骨髓間充質(zhì)干細(xì)胞體外成軟骨分化的影響

【摘要】:目的觀察富血小板血漿(PRP)凝膠及腺病毒介導(dǎo)的骨形態(tài)發(fā)生蛋白2(BMP-2)基因轉(zhuǎn)染對(duì)兔骨髓間充質(zhì)干細(xì)胞(BMSCs)體外成軟骨分化的影響。方法取新西蘭大白兔骨髓培養(yǎng)BMSCs,抽取靜脈血制備PRP,以Ad-GFP-hBMP-2(BMP-2-BMSCs組)及Ad-GFP(對(duì)照組)轉(zhuǎn)染的BMSCs分別與PRP凝膠復(fù)合,繼續(xù)培養(yǎng)。用掃描電鏡、MTT檢測PRP凝膠的生物相容性,RT-PCR檢測各組的Ⅰ型膠原、Ⅱ型膠原、Ⅹ型膠原、蛋白聚糖、SOX-9表達(dá)。結(jié)果掃描電鏡及MTT檢測證實(shí)PRP凝膠具有良好的生物相容性。RT-PCR分析表明,單層培養(yǎng)的BMP-2-BMSCs組的Ⅱ型膠原、蛋白聚糖和SOX-9的表達(dá)水平均高于對(duì)照組(P0.05),但Ⅰ型膠原表達(dá)水平較對(duì)照組明顯下降(P0.05),同時(shí)Ⅹ型膠原表達(dá)水平與對(duì)照組相比差異無統(tǒng)計(jì)學(xué)意義(P0.05);與PRP復(fù)合后,BMP-2-BMSCs組的Ⅱ型膠原、蛋白聚糖和SOX-9表達(dá)水平較對(duì)照組升高(P0.05),而Ⅰ型膠原和Ⅹ型膠原表達(dá)水平與對(duì)照組比較差異無統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論轉(zhuǎn)染BMP-2的BMSCs在PRP凝膠中生長良好,BMP-2基因轉(zhuǎn)染能促進(jìn)BMSCs的體外成軟骨分化。
