【摘要】：目的分析髓核組織高溫相關(guān)絲氨酸蛋白酶A1(HtrA1)的表達水平與椎間盤退變程度(Pfirrmann分級)的相關(guān)性。方法 36例需手術(shù)摘除髓核組織的患者,術(shù)前行磁共振成像檢查,據(jù)矢狀位T2加權(quán)相磁共振對擬手術(shù)節(jié)段予Pfirrmann分級;采用逆轉(zhuǎn)錄PCR及Western blot方法檢測髓核組織中HtrA1的mRNA及蛋白表達水平,對髓核組織HtrA1的表達水平與Pfirrmann等級進行相關(guān)性分析。結(jié)果 36例椎間盤經(jīng)磁共振成像檢查,PfirrmannⅠ級3例、Ⅱ級10例、Ⅲ級11例、Ⅳ級7例、Ⅴ級5例;髓核組織HtrA1 mRNA及蛋白表達水平隨Pfirrmann等級增高而增加,經(jīng)Bonferroni檢驗,除Ⅳ、Ⅴ級間外(P0.05),余各級間兩兩比較差異均有統(tǒng)計學(xué)意義(P0.05),經(jīng)Spearman秩相關(guān)分析:HtrA1 mRNA及蛋白表達水平與Pfirrmann分級間存在秩相關(guān)關(guān)系(P0.000 1)。結(jié)論髓核組織HtrA1 mRNA及蛋白表達水平隨Pfirrmann等級增高而升高,提示HtrA1與椎間盤退變程度存在相關(guān)性。
[Abstract]:Objective to analyze the correlation between the expression of hyperthermia associated serine protease A1 (HtrA1) and the degree of disc degeneration (Pfirrmann grade) in nucleus pulposus. Methods Thirty-six patients with nucleus pulposus were surgically removed. MRI was performed before operation. Pfirrmann was graded according to sagittal T 2-weighted magnetic resonance imaging. The expression of mRNA and protein of HtrA1 in nucleus pulposus was detected by reverse transcription PCR and Western blot, and the correlation between the expression of HtrA1 and Pfirrmann grade in nucleus pulposus was analyzed.
