他克莫司下調(diào)實驗性自身免疫性肌炎小鼠肌組織中IL-17和IL-23 mRNA水平

【摘要】:目的研究白細(xì)胞介素17(IL-17)、IL-23 mRNA在實驗性自身免疫性肌炎(EAM)小鼠肌組織表達(dá)情況,探討他克莫司(TAC)處理對IL-17、IL-23 mRNA水平的影響及對EAM小鼠的治療效果。方法 15只雌性BALB/c小鼠隨機(jī)分為正常對照組、EAM模型組和TAC處理組;HE染色觀察肌組織炎癥程度;實時熒光定量PCR檢測小鼠肌組織IL-17、IL-23 mRNA水平;Pearson法分析二者間的相關(guān)性。結(jié)果與正常組相比,EAM組小鼠肌組織IL-17、IL-23 mRNA水平顯著升高,給予TAC處理后均明顯降低。二者表達(dá)水平與肌組織炎癥評分呈正相關(guān)。與EAM組比較,TAC處理組小鼠肌組織炎癥評分也有所下降。結(jié)論 TAC可下調(diào)EAM小鼠IL-17、IL-23 mRNA水平。
[Abstract]:Objective to study the expression of interleukin 17 (IL-17) and IL-23 mRNA in the muscle tissue of (EAM) mice with experimental autoimmune myositis and to explore the effect of tacrolimus (TAC) treatment on IL-17,IL-23 mRNA level and on EAM mice. Methods Fifteen female BALB/c mice were randomly divided into three groups: normal control group, EAM model group and TAC treated group. The inflammation degree of muscle tissue was observed by HE staining, the IL-17,IL-23 mRNA level of muscle tissue was detected by real-time fluorescence quantitative PCR, and the correlation between them was analyzed by Pearson method. Results compared with the normal group, the level of IL-17,IL-23 mRNA in the EAM group was significantly higher than that in the normal group, and decreased significantly after TAC treatment. There was a positive correlation between the expression level and the inflammatory score of muscle tissue. Compared with EAM group, the inflammatory score of muscle tissue in TAC treated mice was also decreased. Conclusion TAC can down-regulate the level of IL-17,IL-23 mRNA in EAM mice.
