腰椎間盤髓核組織中miR-200c、TrkB表達變化及其與退變程度的相關性

【摘要】:目的觀察微小RNA-200c(miR-200c)與酪氨酸激酶受體B(Trk B)腰椎間盤髓核組織中的表達變化,并探討其與腰椎間盤髓核退變程度的相關性。方法收集腰椎間盤髓核組織標本60例,用Pfirmlann分級對髓核退變程度進行分級,Ⅱ、Ⅲ、Ⅳ級各20例。采用細胞衰老β-半乳糖苷酶實驗檢測不同分級髓核組織中衰老髓核細胞百分比,RT-PCR技術檢測髓核組織中miR-200c、Trk B mRNA表達,Western blotting技術檢測髓核組織中Trk B蛋白表達,采用Pearson相關分析miR-200c、Trk B與衰老髓核細胞的關系。結果 PfirmlannⅡ、Ⅲ、Ⅳ級髓核組織中衰老髓核細胞百分比、miR-200c相對表達量、Trk B mRNA及蛋白相對表達量兩兩比較,P均0.05。miR-200c mRNA與Trk B mRNA相對表達量呈負相關(r=-0.71,P0.05),miR-200c mRNA相對表達量與衰老髓核細胞百分比呈正相關(r=0.75,P0.05),Trk B mRNA相對表達量與衰老髓核細胞百分比呈負相關(r=-0.70,P0.05)。結論腰椎間盤髓核退變程度越高,miR-200c表達越高,Trk B mRNA表達越低;miR-200c可能通過下調Trk B表達參與腰椎間盤髓核組織的退變過程。
