硫化氫對燒傷大鼠體外培養(yǎng)皮膚巨噬細(xì)胞MAPK信號傳導(dǎo)通路中ERK、JNK及p38 mRNA的影響

【摘要】:目的研究H2S對燒傷大鼠體外培養(yǎng)皮膚巨噬細(xì)胞MAPK信號傳導(dǎo)通路中ERK、JNK及p38 m RNA表達水平的影響,從而探究H2S參與創(chuàng)面愈合的分子機制。方法把燒傷大鼠皮膚巨噬細(xì)胞作為研究對象,從30只健康SD大鼠中選取5只大鼠不做任何處理,剩余25只大鼠制造深Ⅱ°燒傷模型(經(jīng)病理切片證實)。從5只正常大鼠中隨機選取1只作為空白對照組,從25只燒傷大鼠中選取12只隨機分為4組:燒傷對照組、Na HS組、格列本脲(GLBN)組、Na HS+格列本脲組,每組3只。造模后分離正;驘齻笫蟊巢科つw基底巨噬細(xì)胞至培養(yǎng)瓶。待穩(wěn)定培養(yǎng)后,分別加入終濃度為200umol/L的相應(yīng)藥物給予干預(yù)(空白對照組和燒傷對照組加入等量的PBS溶液作為對照),在藥物接觸1h、6h、12h每個時相點利用RT-PCR技術(shù)檢測巨噬細(xì)胞中ERK、p38、JNK的m RNA含量變化。結(jié)果1、各組ERK m RNA表達結(jié)果進行比較:(1)燒傷對照組與空白對照組在ERK m RNA轉(zhuǎn)錄水平上有統(tǒng)計學(xué)差異(P0.05);(2)在同一時相點,Na HS組與各組別在ERK m RNA轉(zhuǎn)錄水平上有統(tǒng)計學(xué)差異(P0.05);(3)在同一時相點,Na HS+GLBN組與GLBN組在ERK m RNA轉(zhuǎn)錄水平上統(tǒng)計學(xué)有差異(P0.05);(4)在同一時相點,GLBN組與燒傷組在ERK m RNA轉(zhuǎn)錄水平上比較有差異(P0.05)。2、各組JNK m RNA表達結(jié)果進行兩兩比較:(1)燒傷組與空白對照組在JNK m RNA轉(zhuǎn)錄水平上有統(tǒng)計學(xué)差異(P0.05);(2)在同一時間,Na HS組與各組別在JNK m RNA轉(zhuǎn)錄水平上有差別(P0.05);(3)在同一時相點,Na HS+GLBN組與GLBN組JNK m RNA表達量差別比較有統(tǒng)計學(xué)意義(P0.05);(4)在同一時相點,GLBN組與燒傷組在JNK m RNA轉(zhuǎn)錄水平上比較有差異(P0.05)。3、各組p38 m RNA表達結(jié)果進行比較:(1)燒傷對照組與空白對照組在p38m RNA轉(zhuǎn)錄水平上有統(tǒng)計學(xué)差別(P0.05);(2)在同一時間點,Na HS組與各組別在p38 m RNA轉(zhuǎn)錄水平上有差別(P0.05);(3)在同一時間點,Na HS+GLBN組與GLBN組p38 m RNA表達量差異比較有統(tǒng)計學(xué)意義(P0.05);(4)在同一時間點,GLBN組與燒傷對照組在p38 m RNA轉(zhuǎn)錄水平上差別明顯(P0.05)。結(jié)論硫化氫通過調(diào)控K+通道,使ERK m RNA相對表達水平升高,JNK和p38m RNA表達水平下降,從而影響燒傷大鼠體外培養(yǎng)皮膚巨噬細(xì)胞MAPK信號通路的傳導(dǎo)。
