MMP-2、MMP-3、MMP-9、TIMP-3及Col-Ⅱ因子在兔軟骨細(xì)胞損傷后的表達(dá)變化

【摘要】:檢測(cè)MMP-2、MMP-3、MMP-9、TIMP-3及Ⅱ型膠原細(xì)胞外基質(zhì)因子在正常兔關(guān)節(jié)軟骨細(xì)胞和劃傷后不同時(shí)間點(diǎn)的表達(dá)變化。在無(wú)菌條件下獲取兔膝關(guān)節(jié)軟骨細(xì)胞,原代體外培養(yǎng)軟骨細(xì)胞,六孔板內(nèi)制備細(xì)胞損傷模型。用顯微鏡觀察正常細(xì)胞和損傷后1 d、3 d和7 d 3個(gè)時(shí)間點(diǎn)的軟骨細(xì)胞增殖情況。采用實(shí)時(shí)PCR檢測(cè)正常軟骨細(xì)胞和損傷后1 d、3 d和7 d 3個(gè)時(shí)間點(diǎn)的基質(zhì)金屬蛋白酶-2、3和9,基質(zhì)金屬蛋白酶抑制物-3及Ⅱ型膠原的m RNA的表達(dá)水平。成功分離軟骨,經(jīng)原代培養(yǎng)后成功建立損傷細(xì)胞模型。MMP-2損傷后第1天與正常組相比升高,第3天時(shí)下降,第7天明顯水平最高。與正常組相比,MMP-3在細(xì)胞損傷后第1天表達(dá)明顯下降,隨后第3天逐漸下降,第7天與第3天未見明顯變化。MMP-9劃傷后第1天比正常組升高,第3天上升至最高水平,第7天下降。TIMP-3基因在損傷后第1天與正常組相比明顯下降,隨后第3天稍有升高,第7天水平最低。Ⅱ型膠原劃傷后第1天與正常組相比明顯升高,隨后第3天呈下降趨勢(shì)接近于正常組,第7天又呈上升趨勢(shì)。MMP-2、MMP-3、MMP-9、TIMP-3因子和Ⅱ型膠原在細(xì)胞損傷后的不同時(shí)間點(diǎn)有不同的表達(dá)行為,這可為調(diào)節(jié)細(xì)胞外基質(zhì)基因治療關(guān)節(jié)軟骨損傷選擇合適的靶基因和時(shí)間窗提供實(shí)驗(yàn)依據(jù)。
