尼莫地平復(fù)合7.5%高滲鹽水對七氟醚誘導(dǎo)老齡大鼠海馬神經(jīng)元凋亡的影響

【摘要】:目的探討尼莫地平復(fù)合7.5%高滲鹽水對七氟醚誘導(dǎo)老齡大鼠海馬神經(jīng)元凋亡的影響。方法健康雄性Wistar大鼠96只,18月齡,體重450~500g,采用隨機(jī)數(shù)字表法將其分為四組(n=24):對照組(C組)、尼莫地平組(N組)、高滲鹽水組(HS組)和尼莫地平+高滲鹽水組(NHS組)。C組腹腔和尾靜脈注射生理鹽水;N組腹腔注射尼莫地平1 mg/kg,尾靜脈注射生理鹽水;HS組尾靜脈注射7.5%高滲鹽水4 ml/kg,腹腔注射生理鹽水;NHS組腹腔注射尼莫地平1mg/kg,尾靜脈注射7.5%高滲鹽水4ml/kg。30min后四組大鼠吸入3%七氟醚2h。于麻醉前1d、麻醉后1、3、7d行Morris水迷宮實(shí)驗(yàn),實(shí)驗(yàn)結(jié)束后每組隨機(jī)處死8只大鼠,取海馬組織,采用流式細(xì)胞術(shù)測定海馬神經(jīng)元凋亡率和胞漿鈣離子濃度;采用RT-PCR法測定海馬Bcl-2、Bax的mRNA表達(dá)量。結(jié)果與C組比較,N、HS、NHS組大鼠麻醉后1、3、7d逃避潛伏期明顯縮短,穿越原平臺(tái)次數(shù)明顯增加,麻醉后1、7d海馬神經(jīng)元凋亡率和胞漿鈣離子濃度明顯減低,Bcl-2 mRNA表達(dá)量明顯上調(diào),Bax mRNA表達(dá)量明顯下調(diào),Bax/Bcl-2比值明顯降低(P0.05);與NHS組比較,N和HS組大鼠麻醉后1、3、7d逃避潛伏期明顯延長,穿越原平臺(tái)次數(shù)明顯減少,麻醉后1、7d海馬神經(jīng)元凋亡率和胞漿鈣離子濃度明顯增高,Bcl-2 mRNA表達(dá)量明顯下調(diào),Bax mRNA表達(dá)量明顯上調(diào),Bax/Bcl-2比值明顯升高(P0.05)。結(jié)論尼莫地平復(fù)合7.5%高滲鹽水可抑制鈣超載,降低七氟醚誘導(dǎo)老齡大鼠海馬神經(jīng)元凋亡率,且其作用優(yōu)于單獨(dú)給藥。
[Abstract]:Aim to investigate the effect of nimodipine combined with 7.5% hypertonic saline on the apoptosis of hippocampal neurons induced by sevoflurane in aged rats. Methods 96 healthy male Wistar rats were 18 months old. The body weight was 450 ~ 500g. It was randomly divided into four groups: control group (group C), nimodipine group (group N), hypertonic saline group (HS group) and nimodipine hypertonic saline group (NHS group). C group was injected intraperitoneally and caudal intravenously with normal saline. One mg / kg nimodipine was injected intravenously, 7.5% hypertonic saline 4 ml / kg was injected into caudal vein of HS group, 1 mg / kg nimodipine was injected intraperitoneally in NHS group, and 3% sevoflurane was inhaled by caudal vein after 7.5% hypertonic saline 4ml/kg.30min. Morris water maze test was performed 1 day before anesthesia and 1 day 3 days after anesthesia. Eight rats in each group were randomly killed at the end of the experiment. The hippocampal tissue was taken out. The apoptosis rate of hippocampal neurons and the concentration of Ca ~ (2 +) in hippocampal neurons were measured by flow cytometry. The expression of Bax mRNA in hippocampus was measured by RT-PCR. Results compared with group C, the escape latency of NHS group was significantly shorter than that of group C, and the times of crossing the original platform increased significantly. The apoptotic rate of hippocampal neurons and cytoplasmic calcium concentration significantly decreased the expression of Bcl 2 mRNA on the 1st and 7th day after anesthesia, the expression of Bax mRNA decreased significantly (P0.05), and the escape latency of rats in the N and HS groups was significantly longer than that in the NHS group on the 1st and 3rd day after anesthesia, and the expression of Bax mRNA was significantly down-regulated (P0.05), and the escape latency of the rats in the N and HS groups was significantly longer than that in the NHS group. The frequency of traversing the original platform was significantly decreased, and the apoptotic rate of hippocampal neurons and the concentration of Ca ~ (2 +) in the hippocampus were significantly increased on the 1st and 7th day after anesthesia. The expression of Bax mRNA was down-regulated and the ratio of Bax / Bax mRNA was upregulated significantly (P0.05). Conclusion Nimodipine combined with 7.5% hypertonic saline can inhibit calcium overload and decrease apoptosis rate of hippocampal neurons induced by sevoflurane in aged rats.
