TNF-α在構建大鼠骨性關節(jié)炎模型中對內(nèi)質(zhì)網(wǎng)應激的影響探討

【摘要】:目的觀察不同劑量腫瘤壞死因子-α(tumor necrosis factor,TNF-α)在構建大鼠骨性關節(jié)炎(osteoarthritis,OA)模型中對內(nèi)質(zhì)網(wǎng)應激的影響,初步探討大鼠骨性關節(jié)炎病理過程中內(nèi)質(zhì)網(wǎng)應激的作用。方法將20只SPF級SD雄性大鼠隨機分為健康對照組、TNF-α1ng/m L干預組、TNF-α5ng/m L干預組和TNF-α10ng/m L干預組,分別給予不同劑量TNF-α處理,4周后取材,培養(yǎng)提取的各組原代軟骨細胞,采用MTT法檢測不同劑量TNF-α對軟骨細胞活力的影響;ELISA法檢測內(nèi)皮細胞粘附分子-1(endothelial cell adhesion molecule-1,s ICAM-1)含量變化,TUNEL法檢測各組軟骨細胞凋亡率,蛋白印跡法(Western blot)檢測炎癥因子MMP9、ICAM1的表達以及內(nèi)質(zhì)網(wǎng)應激指標IRE1、p-IRE1、GRP78的水平變化。結果 MTT法檢測提示加入不同劑量的TNF-α后軟骨細胞活力未見明顯改變;TUNEL法檢測結果提示,與對照組相比,TNF-α干預組軟骨細胞凋亡率較高(P0.01),并呈濃度依賴性促進細胞凋亡;Western blot結果表明,與對照組相比,軟骨細胞的p-IRE1、IRE1、GRP78、MMP9及ICAM1的表達水平呈TNF-α濃度依賴性升高(P0.01)。結論在TNF-α構建大鼠骨性關節(jié)炎模型中,炎癥因子與內(nèi)質(zhì)網(wǎng)應激指標均發(fā)生顯著改變,內(nèi)質(zhì)網(wǎng)應激可能在大鼠骨性關節(jié)炎的發(fā)生發(fā)展過程中發(fā)揮重要影響,且當TNF-α濃度為10 ng/m L時,其對炎性因子及內(nèi)質(zhì)網(wǎng)指標的作用效果最為顯著。
[Abstract]:Objective to observe the effects of different doses of tumor necrosis factor- 偽 on endoplasmic reticulum stress in rat osteoarthritis model, and to explore the role of endoplasmic reticulum stress in the pathological process of rat osteoarthritis. Methods Twenty Sprague-SPF male rats were randomly divided into two groups: TNF- 偽 1ng/m L intervention group and TNF- 偽 10ng/m L intervention group. The primary chondrocytes of each group were cultured after 4 weeks of treatment with different doses of TNF- 偽 10ng/m. The effect of different doses of TNF- 偽 on chondrocyte viability was detected by MTT assay. The changes of the content of endothelial cell adhesion molecule cell adhesion molecule-1and ICAM-1 were detected by Elisa. The apoptosis rate of chondrocytes in each group was detected by Tunel method. Western blot was used to detect the expression of ICAM1 and the level of endoplasmic reticulum stress (IRE1) -IRE1 / GRP78. Results MTT assay showed that the activity of chondrocytes did not change after adding different doses of TNF- 偽. Compared with the control group, the apoptotic rate of chondrocytes in the TNF- 偽 intervention group was higher than that in the control group. The results showed that the expression of p-IRE1, IRE1, GRP78, MMP9 and ICAM1 in chondrocytes was increased in a concentration-dependent manner in a concentration dependent manner (P0.01a), and the results showed that the expression levels of p-IRE1, IRE1, GRP78, MMP9 and ICAM1 in chondrocytes were increased in a concentration-dependent manner. Conclusion the inflammatory factors and endoplasmic reticulum stress were significantly changed in rat osteoarthritis induced by TNF- 偽. Endoplasmic reticulum stress may play an important role in the development of osteoarthritis in rats. When the concentration of TNF- 偽 was 10 ng/m / L, the effect of TNF- 偽 on inflammatory factors and endoplasmic reticulum was the most significant.
