水溫改變對(duì)斑馬魚脊髓損傷修復(fù)的影響

【摘要】:目的利用斑馬魚脊髓損傷模型,研究水溫改變對(duì)脊髓損傷修復(fù)的影響,并檢測(cè)在脊髓修復(fù)過程中,損傷處細(xì)胞數(shù)目以及腦內(nèi)相關(guān)基因表達(dá)的變化情況。方法利用手術(shù)構(gòu)建斑馬魚脊髓損傷模型;通過改變斑馬魚生活的水溫來觀察其脊髓損傷后游動(dòng)能力的變化情況;用振動(dòng)切片技術(shù)及免疫組織化學(xué)觀察脊髓損傷處不同時(shí)間點(diǎn)細(xì)胞數(shù)目的改變情況;實(shí)時(shí)熒光定量PCR法分析腦內(nèi)相關(guān)基因gdnf、nos的表達(dá)差異。結(jié)果 28℃、30℃、32℃水溫均不會(huì)導(dǎo)致未手術(shù)組與假手術(shù)組斑馬魚游動(dòng)能力的改變(P0.05);32℃水溫環(huán)境能夠使脊髓損傷組斑馬魚比28℃、30℃水溫環(huán)境的斑馬魚恢復(fù)游動(dòng)的能力明顯增強(qiáng)(P0.05)。脊髓切片染色實(shí)驗(yàn)結(jié)果顯示,脊髓損傷后損傷處細(xì)胞數(shù)目明顯增多(P0.05)。相同時(shí)間點(diǎn),水溫為32℃比28℃、30℃細(xì)胞數(shù)目增多更加明顯(P0.05)。實(shí)時(shí)熒光定量實(shí)驗(yàn),gdnf基因在脊髓損傷術(shù)后與對(duì)照組相比在24 h、7 d、14 d均明顯升高(P0.05);nos基因在脊髓損傷術(shù)后24 h明顯升高(P0.05),7 d時(shí)與相同時(shí)間點(diǎn)假手術(shù)組相比無明顯差異(P0.05),而14 d時(shí)與相同時(shí)間點(diǎn)假手術(shù)組相比明顯降低(P0.05)。結(jié)論適當(dāng)提高水環(huán)境溫度可促進(jìn)斑馬魚脊髓損傷后恢復(fù)。
[Abstract]:Objective to study the effect of water temperature on the repair of spinal cord injury in zebrafish, and to detect the changes of the number of cells and the expression of related genes in the brain during spinal cord repair. Methods the spinal cord injury model of zebrafish was established by operation, and the swimming ability of zebrafish was observed by changing the water temperature of zebrafish life.
