【摘要】：目的 1、分析CREPT在腹膜后脂肪肉瘤中表達(dá)及對(duì)預(yù)后的影響;2、分析CREPT對(duì)SW872細(xì)胞增殖能力的影響。方法 1、收集2007年6月至2014年3月我院收治的腹膜后脂肪肉瘤患者的臨床資料和腫瘤蠟塊,并對(duì)患者預(yù)后進(jìn)行隨訪。通過病理蠟塊連續(xù)切片HE染色和免疫組化染色,分析CREPT在脂肪肉瘤的表達(dá)及預(yù)后的影響。2、采用RNA干擾(RNAi)技術(shù)將CREPT進(jìn)行表達(dá)抑制,通過MTT法、細(xì)胞平板克隆實(shí)驗(yàn)、劃痕實(shí)驗(yàn)檢測CREPT對(duì)SW872細(xì)胞增殖的影響。結(jié)果CREPT在脂肪肉瘤具有高表達(dá),其表達(dá)強(qiáng)弱與腫瘤的級(jí)別(χ2=67.070,P0.01)、核分裂數(shù)(χ2=52.055,P0.01)、腫瘤壞死(χ2=25.546,P0.01)、腫瘤分化(χ2=57.109,P0.01)明顯相關(guān),Log Rank檢驗(yàn)發(fā)現(xiàn),CREPT表達(dá)強(qiáng)弱(χ2=37.759,P0.01),核分裂數(shù)(χ2=71.644,P0.01),腫瘤分化(χ2=30.357,P0.01),腫瘤壞死(χ2=27.369,P0.01),腫瘤級(jí)別(χ2=51.374,P0.01),手術(shù)方式(χ2=11.178,P0.01)是影響患者生存時(shí)間的主要因素。Cox多因素生存分析顯示CREPT表達(dá)(RR=2.289,P=0.015)和核分裂(RR=2.444,P=0.005)數(shù)是預(yù)后的獨(dú)立影響因素。通過MTT、平板克隆實(shí)驗(yàn)、劃痕實(shí)驗(yàn)證明CREPT對(duì)SW872細(xì)胞增殖具有促進(jìn)作用,抑制CREPT表達(dá),細(xì)胞增殖能力減弱(P0.05)。結(jié)論CREPT在脂肪肉瘤中高度表達(dá),且與預(yù)后明顯相關(guān)。抑制CREPT表達(dá)，SW872細(xì)胞增殖能力明顯減弱。
[Abstract]:Objective 1. To analyze the expression of CREPT in retroperitoneal liposarcoma and its influence on prognosis, and to analyze the effect of CREPT on the proliferation of SW872 cells. Methods 1. The clinical data and tumor wax of retroperitoneal liposarcoma treated in our hospital from June 2007 to March 2014 were collected and the prognosis was followed up. The expression and prognosis of CREPT in liposarcoma were analyzed by HE staining and immunohistochemical staining. The expression of CREPT was inhibited by RNA interference technique. The expression of CREPT was inhibited by MTT method. The effect of CREPT on proliferation of SW872 cells was detected by scratch test.
