高頻正弦波振動抑制廢用性肌萎縮的凋亡進(jìn)程

【摘要】:目的:探討高頻正弦波振動(HFV)對骨骼肌在廢用狀態(tài)時細(xì)胞凋亡的對抗作用。方法:建立廢用動物模型,改進(jìn)振動儀的關(guān)鍵技術(shù),采用短時間歇式振動的方法,結(jié)合電鏡觀察、免疫組化染色法和TUNEL法檢測,觀察HFV對制動大鼠比目魚肌(SOL)濕重體重比、肌纖維橫截面積、肌纖維超微結(jié)構(gòu)的變化、Bcl-2和Bax的表達(dá)情況。結(jié)果:(1)制動后大鼠SOL濕重體重比為30.22±2.68,下降了36.43%。肌纖維的橫截面積為3789.56±95.14μm2,下降了37.77%。HFV組相對制動組的SOL濕重體重比及橫截面積均有所增加(P0.05);(2)電鏡可見制動后梭外肌病變明顯,核內(nèi)染色質(zhì)團(tuán)塊狀凝集。梭內(nèi)肌部分肌膜溶解消失,細(xì)胞核變形。振動組的梭外肌Z線結(jié)構(gòu)比較清晰,梭內(nèi)肌纖維結(jié)構(gòu)未見明顯改變;(3)制動后大鼠SOL中Bcl-2陽性表達(dá)率為20%,明顯低于對照組的表達(dá)情況(P0.05)。制動后Bax陽性表達(dá)率為80%,明顯高于對照組的表達(dá)情況(P0.05)。制動后AI為20%,明顯高于對照組(P0.05)。HFV組相對制動組的Bcl-2陽性表達(dá)率均有所增加,Bax陽性表達(dá)率以及AI有所降低(P0.05)。結(jié)論:改進(jìn)后的抗肌萎縮振動儀操作性較強(qiáng),對制動大鼠廢用性肌萎縮的凋亡進(jìn)程有抑制作用。
[Abstract]:Aim: to investigate the effect of high frequency sine wave vibration (HFV) on apoptosis of skeletal muscle. Methods: to establish a waste animal model, improve the key technology of vibrator, observe the wet weight / body weight ratio of HFV to soleus muscle of brake rats by using the method of short-term intermittent vibration, electron microscope observation, immunohistochemical staining and TUNEL method. The cross sectional area of muscle fiber and the changes of ultrastructure of muscle fiber were detected by the expression of Bcl-2 and Bax. Results the wet weight / body weight ratio of SOL was 30.22 鹵2.68, which decreased by 36.43%. The cross sectional area of muscle fiber was 3789.56 鹵95.14 渭 m ~ (2), which decreased the SOL wet weight / body weight ratio and cross-sectional area of relative immobilization group in 37.77%.HFV group. Part of the intrafusal muscle membrane dissolves and the nucleus deforms. In vibration group, the Z line structure of extrafusal muscle was clear, and the fiber structure of intrafusal muscle did not change significantly. The positive expression rate of Bcl-2 in SOL of rats after immobilization was 20%, which was significantly lower than that of control group (P 0.05). The positive expression rate of Bax after immobilization was 80%, which was significantly higher than that of control group (P 0.05). After immobilization, AI was 20%, which was significantly higher than that in the control group (P 0.05). The positive expression rate of Bcl-2 in the control group was higher than that in the control group. The positive expression rate of Bax was increased and the AI was decreased (P 0.05). Conclusion: the improved anti-atrophy vibrator has strong operability and can inhibit the process of apoptosis of disused muscle atrophy in immobilization rats.
