【摘要】：目的觀察拉喹莫德灌胃對(duì)大鼠腰椎間盤退變的治療作用。方法將60只大鼠隨機(jī)分為空白對(duì)照組(A組)、模型對(duì)照組(B組)、拉喹莫德組(C組)各20只。B、C組建立腰椎間盤退變模型,術(shù)后2周C組給予拉喹莫德按60 mg/(kg·d)灌胃,A、B組予以等體積的生理鹽水灌胃,連續(xù)6周持續(xù)給藥。第8周處死各組大鼠,手術(shù)取出L_(4~5)節(jié)段腰椎間盤中的髓核組織,采用光鏡和電鏡觀察大鼠椎間盤髓核組織病理,采用Western blot法檢測(cè)大鼠椎間盤髓核組織中HIF-2α。結(jié)果 C組較B組退變壞死細(xì)胞數(shù)量明顯減少,脊索細(xì)胞數(shù)目較多僅少量細(xì)胞核遭到破壞,膠原纖維和周期性橫紋大部分可見且有序。A、B、C組大鼠椎間盤髓核組織中HIF-2α相對(duì)表達(dá)量分別為115.6±12.5、150.3±18.2、127.2±18.7,兩兩相比,P均0.05。結(jié)論拉喹莫德灌胃對(duì)大鼠腰椎間盤退變具有治療作用,這可能是通過減少髓核組織中HIF-2α的表達(dá)實(shí)現(xiàn)的。
[Abstract]:Objective to observe the therapeutic effect of laquimod on lumbar disc degeneration in rats. Methods Sixty rats were randomly divided into three groups: control group A (n = 20), model control group B (n = 20), laquimod group C (n = 20) and control group C (n = 20). Two weeks after operation, group C was given the same volume of normal saline by intragastric administration of laquimod for 60 mg/(kg d, and the drug was given continuously for 6 weeks. At the 8th week, the rats in each group were killed, and the nucleus pulposus tissue was removed from the lumbar intervertebral disc of L _ (4 / 5) segment by operation. The histopathology of the nucleus pulposus was observed by light microscope and electron microscope, and the HIF-2 偽 in the nucleus pulposus tissue of the rat disc was detected by Western blot method. Results the number of degenerative necrotic cells in group C was significantly lower than that in group B, and the number of notochord cells was more than that in group B. The relative expression of HIF-2 偽 was 115.6 鹵12.5150.3 鹵18.2127.2 鹵18.7 in the nucleus pulposus of the intervertebral disc in most of the collagenous fibers and periodic transverse striae, and the relative expression of HIF-2 偽 in the nucleus pulposus was 115.6 鹵12.5150.3 鹵18.2127.2 鹵18.7in the control group (P < 0.05). Conclusion Laquimod has therapeutic effect on the degeneration of lumbar intervertebral disc in rats, which may be achieved by reducing the expression of HIF-2 偽 in nucleus pulposus.
