塞來昔布對(duì)人成骨細(xì)胞增殖的影響及其作用機(jī)制

【摘要】:目的觀察塞來昔布對(duì)人成骨細(xì)胞增殖的影響及其作用機(jī)制。方法將人成骨細(xì)胞分為3個(gè)組:空白對(duì)照組、IL-1刺激組(陰性對(duì)照組)、塞來昔布+IL-1刺激組(藥物干預(yù)組),用RT-PCR法和Western blot法分別檢測(cè)三組成骨細(xì)胞環(huán)氧化酶-2(COX-2)、膜相關(guān)前列腺素E2合成酶1(m PGES-1)mRNA和蛋白的表達(dá)。MTT法檢測(cè)塞來昔布對(duì)陰性對(duì)照組人成骨細(xì)胞增殖的影響。結(jié)果 RT-PCR法和Western blot法實(shí)驗(yàn)結(jié)果顯示,同空白對(duì)照組相比,陰性對(duì)照組的人成骨細(xì)胞中COX-2、m PGES-1 mRNA和蛋白的表達(dá)均增加;同陰性對(duì)照組相比,藥物干預(yù)組的COX-2、m PGES-1mRNA和蛋白的表達(dá)均降低。塞來昔布對(duì)IL-1刺激的人成骨細(xì)胞的生長具有抑制作用,呈現(xiàn)劑量依賴性,濃度越大,抑制作用越大;并且隨著作用時(shí)間的延長,細(xì)胞的抑制作用增大。結(jié)論塞來昔布可以抑制炎性狀態(tài)下人成骨細(xì)胞的生長,可能是通過調(diào)控COX-2、m PGES-1 mRNA和蛋白的表達(dá),使其表達(dá)下調(diào)而發(fā)揮作用。
[Abstract]:Objective to observe the effect of celecoxib on the proliferation of human osteoblasts and its mechanism. Methods Human osteoblasts were divided into three groups: blank control group (negative control group) and celecoxib IL-1 stimulation group (Celecoxib IL-1 stimulation group). The membrane phase of osteoblasts was detected by RT-PCR method and Western blot method. The effect of celecoxib on the proliferation of human osteoblasts in negative control group was detected by MTT assay. Results the results of RT-PCR assay and Western blot assay showed that the expression of COX-2 m PGES-1 mRNA and protein in osteoblasts in negative control group was higher than that in blank control group, but the expression of COX-2 m PGES-1mRNA and protein in drug intervention group was lower than that in negative control group. Celecoxib inhibited the growth of human osteoblasts stimulated by IL-1 in a dose-dependent manner. Conclusion Celecoxib can inhibit the growth of human osteoblasts in inflammatory state, which may play a role by regulating the expression of COX-2 PGES-1 mRNA and protein and down-regulating the expression of celecoxib.
