甲狀旁腺激素(1-34)對(duì)骨質(zhì)疏松相關(guān)性骨性關(guān)節(jié)炎的作用

【摘要】:目的本次實(shí)驗(yàn)的目的旨在應(yīng)用甲狀旁腺激素(Parathyroid hormone,PTH)干預(yù)Sprague-Dawley(SD)大鼠雙側(cè)卵巢切除術(shù)(Ovariectomy,OVX)導(dǎo)致的骨質(zhì)疏松相關(guān)的骨關(guān)節(jié)炎(Osteoarthritis,OA)模型中,研究PTH對(duì)關(guān)節(jié)軟骨及軟骨下骨的作用。方法18只6月齡SPF級(jí)SD大鼠隨機(jī)分為3組每組6只,即:Sham組、OVX+NS組和OVX+PTH組,OVX+NS組和OVX+PTH組行雙側(cè)卵巢切除術(shù)。術(shù)后一周OVX+PTH組給予PTH10mg/kg/day皮下注射,每周五次,OVX+NS組給予等量生理鹽水皮下注射。給藥12周后處死動(dòng)物,對(duì)膝關(guān)節(jié)行Micro-CT骨組織形態(tài)學(xué)測(cè)定,甲苯胺藍(lán)染色和COL-II、MMP-13免疫組化染色。甲苯胺藍(lán)染色切片行OARSI組織學(xué)評(píng)分;免疫組化陽性蛋白表達(dá)行IOD評(píng)分。應(yīng)用SPSS17對(duì)各組數(shù)據(jù)進(jìn)行統(tǒng)計(jì)學(xué)分析。結(jié)果1在OARSI組織學(xué)評(píng)分中OVX+NS組與OVX+PTH組比較,OVX+NS組與Sham組比較,總軟骨退變寬度評(píng)分均無明顯差異,未出現(xiàn)明顯軟骨損傷,也未出現(xiàn)明顯基質(zhì)和軟骨淺層細(xì)胞損失,潮線連續(xù)性好,關(guān)節(jié)邊緣未見明顯骨贅形成。2 Micro-CT分析:OVX+NS組與Sham組比較,OVX+NS組BMD顯著降低(P0.05),OVX+PTH組與OVX+NS組比較,OVX+PTH組BMD顯著增加(P0.05)。Sham組與OVX+NS組比較,OVX+NS組骨體積分?jǐn)?shù)(BV/TV)顯著降低(P0.05);OVX+NS組與OVX+PTH組相比較,OVX+PTH組骨體積分?jǐn)?shù)(BV/TV)、骨小梁厚度(Tb.Th)均顯著升高(P0.05)。3關(guān)節(jié)軟骨免疫組化IOD評(píng)分顯示:OVX+PTH組與OVX+NS組比較,OVX+PTH組Col-II陽性表達(dá)有增加趨勢(shì),但未見顯著差異;OVX+NS組與Sham組比較,OVX+NS組Col-II陽性表達(dá)顯著降低,有統(tǒng)計(jì)學(xué)意義差異(P0.05)。OVX+NS組分別與Sham組和OVX+PTH組比較,OVX+NS組MMP-13免疫組化染色陽性表達(dá)均顯著增加(P0.05)。結(jié)論1.OVX加速大鼠軟骨下骨和關(guān)節(jié)軟骨退變,6月齡OVX大鼠是一種輕微的絕經(jīng)后OA大鼠模型。2.PTH可改善OVX導(dǎo)致的軟骨下骨骨量丟失與微結(jié)構(gòu),這可能有助于維持關(guān)節(jié)軟骨的局部力學(xué)環(huán)境。3.PTH減緩關(guān)節(jié)軟骨退變。
[Abstract]:Objective to investigate the effects of PTH on articular cartilage and subchondral bone in bilateral ovariectomy-induced osteoarthritis induced by bilateral ovariectomy (OVX) in rats with parathyroid hormone (PTH). Methods Eighteen 6-month-old SPF SD rats were randomly divided into 3 groups, 6 rats in each group, namely, the OVX PTH group, the OVX PTH group, the OVX NS group and the OVX PTH group. One week after operation, the OVX PTH group was subcutaneously injected with PTH10mg/kg/day, and the OVX NS group was given the same amount of normal saline subcutaneously five times a week. After 12 weeks of administration, the animals were killed. The bone morphology of Micro-CT, toluidine blue staining and COL-IIP MMP-13 immunohistochemical staining were performed on the knee joint. Toluidine blue staining section was stained with OARSI histological score, and immunohistochemical positive protein expression was evaluated with IOD score. The data of each group were analyzed statistically by SPSS17. Results (1) in OARSI histological score, there was no significant difference in total cartilage degeneration width between OVX NS group and OVX PTH group, and there was no obvious cartilage injury, and no obvious matrix and superficial cartilage cell loss in OVX NS group and Sham group. The continuity of the tidal line is good. 2 Micro-CT analysis of osteophyte formation at articular edge; BMD in OVX NS group was significantly lower than that in Sham group; BMD in OVX PTH group was significantly lower than that in OVX NS group; BMD in OVX PTH group was significantly higher than that in OVX NS group and the bone volume fraction in OVX NS group was significantly lower than that in OVX NS group. Compared with OVX PTH group, the bone volume fraction (BV / TVV) and trabecular thickness (Tb. Th) of OVX PTH group were significantly higher than that of OVX PTH group. The immunohistochemical IOD score of articular cartilage showed that the Col-II positive expression of OVX PTH group in the PTH group was increased compared with that in the OVX NS group. However, there was no significant difference between OVX NS group and Sham group. The positive expression of MMP-13 in OVX NS group was significantly lower than that in Sham group and OVX PTH group. There was a significant difference in Col-II expression between OVX NS group and Sham group and OVX PTH group. The expression of MMP-13 immunohistochemical staining in OVX NS group was significantly higher than that in Sham group and OVX PTH group. Conclusion 1.OVX accelerated subchondral bone and articular cartilage degeneration in 6-month-old OVX rats is a mild postmenopausal OA rat model. 2. PTH can improve the bone loss and microstructure of subchondral bone induced by OVX. This may help to maintain the local mechanical environment of articular cartilage. 3. PTH can slow the degeneration of articular cartilage.
