人臍帶間充質(zhì)干細(xì)胞聯(lián)合富含血小板血漿促進(jìn)大鼠跟腱損傷的修復(fù)

【摘要】:目的探討不同濃度富含血小板血漿(platelet-rich plasma,PRP)在體外對(duì)人臍帶間充質(zhì)干細(xì)胞(human umbilical cord mesenchymal stem cells,h UC-MSCs)增殖、分化的影響,評(píng)估h UC-MSCs聯(lián)合PRP對(duì)大鼠跟腱損傷修復(fù)的影響。方法 PRP活化后成為PRP-clot releasate(PRCR)。取P3代h UC-MSCs于培養(yǎng)皿中培養(yǎng)24 h后,分別加入含0%(對(duì)照組)、10%、20%、40%PRCR的DMEM/F12培養(yǎng)基,于第1、2、3、4天測(cè)定h UC-MSCs的增殖速度。P3代h UC-MSCs按相同分組培養(yǎng),于第1、3、5天收集細(xì)胞并提取蛋白。Western blot檢測(cè)h UC-MSCs中成肌腱分化標(biāo)志性基因肌腱蛋白C(tenascin c,TNC)、SCX(scleraxis)、Ⅰ型膠原的表達(dá)。選取48只8周齡雄性SD大鼠,Ⅰ型膠原酶溶液50μL注射于大鼠右側(cè)跟腱制備跟腱損傷模型,3 d后于損傷部位再次分別注射50μL生理鹽水(對(duì)照)、h UC-MSCs懸液、PRP或PRP+h UC-MSCs混懸液,術(shù)后2、4周安樂死大鼠,Western blot檢測(cè)跟腱中TNC、SCX及Ⅰ型膠原的表達(dá),跟腱切片HE染色行組織學(xué)評(píng)估。結(jié)果 PRCR在體外對(duì)h UC-MSCs增殖均有促進(jìn)作用,20%PRCR組促增殖作用最顯著(P0.05);20%PRCR組較對(duì)照組在體外可以顯著地促進(jìn)h UCMSCs成肌腱分化標(biāo)志性基因的表達(dá)(P0.05);術(shù)后2、4周,PRP+h UC-MSCs組跟腱中成肌腱分化相關(guān)基因在蛋白水平表達(dá)更高,跟腱切片HE染色顯示PRP+h UC-MSCs組纖維排列更規(guī)整,炎細(xì)胞更少。結(jié)論適宜濃度PRP在體外顯著促進(jìn)了h UC-MSCs的增殖、成肌腱分化;PRP聯(lián)合h UC-MSCs能有效促進(jìn)大鼠受損跟腱的修復(fù)。
[Abstract]:Objective to investigate the effects of platelet-rich plasma-rich plasma with different concentrations of platelet-rich plasma on proliferation and differentiation of human umbilical cord mesenchymal stem cells hUC-MSCs in vitro, and to evaluate the effects of h UC-MSCs combined with PRP on the repair of Achilles tendon injury in rats. Methods PRP was activated to become PRP-clot release. After cultured in culture dish for 24 hours, P3 generation h UC-MSCs was added into DMEM/F12 medium containing 0 (control group 10) and 20% 40 PRCR respectively. The proliferation rate of h UC-MSCs.
