植入含感覺神經(jīng)束和血管束組織工程骨修復(fù)股骨大段骨缺損:降鈣素基因相關(guān)肽Ⅰ型受體的中長期表達(dá)

【摘要】:背景:前期實(shí)驗(yàn)證實(shí),植入含血管束或感覺神經(jīng)的組織工程骨促成骨程度與其內(nèi)的感覺神經(jīng)肽受體表達(dá)相關(guān)。目的:觀察植入含感覺神經(jīng)束和血管束的組織工程骨修復(fù)兔股骨大段骨缺損,對(duì)降鈣素基因相關(guān)肽Ⅰ型受體中長期表達(dá)的影響。方法:取36只新西蘭大白兔,制作單側(cè)股骨1.5 cm缺損模型,隨機(jī)分3組,感覺神經(jīng)組于骨缺損處植入成骨誘導(dǎo)7 d的骨髓間充質(zhì)干細(xì)胞-β-磷酸三鈣組織工程骨,并在組織工程骨側(cè)槽內(nèi)植入自體感覺神經(jīng)束;血管束組于骨缺損處植入成骨誘導(dǎo)7 d的骨髓間充質(zhì)干細(xì)胞-β-磷酸三鈣組織工程骨,并在組織工程骨側(cè)槽內(nèi)植入自體股血管束;空白對(duì)照組于骨缺損處植入成骨誘導(dǎo)7 d的骨髓間充質(zhì)干細(xì)胞-β-磷酸三鈣組織工程骨。植入后24,48周,進(jìn)行X射線、免疫組織學(xué)及熒光定量PCR檢測(cè)。結(jié)果與結(jié)論:1X射線檢查:感覺神經(jīng)組、血管束組X射線評(píng)分高于空白對(duì)照組(P0.05),感覺神經(jīng)組、血管束組X射線評(píng)分比較差異無顯著性意義;2熒光定量PCR檢測(cè):血管束組降鈣素基因相關(guān)肽Ⅰ型受體表達(dá)量高于感覺神經(jīng)組、空白對(duì)照組(P0.05),感覺神經(jīng)組與空白對(duì)照組比較差異無顯著性意義;3免疫組織化學(xué)觀察:感覺神經(jīng)組、血管束組降鈣素基因相關(guān)肽Ⅰ型受體陽性表達(dá)強(qiáng)于空白對(duì)照組;4結(jié)果表明:植入含血管束的組織工程骨可促進(jìn)降鈣素基因相關(guān)肽Ⅰ型受體的表達(dá)。
[Abstract]:Background: previous studies have demonstrated that the degree of bone promotion by implanting tissue engineered bone containing vascular bundles or sensory nerves is related to the expression of sensory neuropeptide receptors. Aim: to investigate the effect of implanted tissue engineered bone containing sensory nerve bundle and vascular bundle on the long-term expression of calcitonin gene-related peptide type I receptor in rabbit femur. Methods: 36 New Zealand white rabbits were randomly divided into 3 groups. The sensory nerve group implanted bone marrow mesenchymal stem cells (BMSCs)-尾 -tricalcium phosphate tissue engineering bone at the site of bone defect for 7 days. Autogenous sensory nerve bundle was implanted into the lateral groove of tissue engineering bone, and bone marrow mesenchymal stem cell-尾-tricalcium phosphate tissue engineering bone was implanted in the blood tube bundle group at the site of bone defect for 7 days, and autogenous femoral vascular bundle was implanted in the tissue engineering bone side groove. Bone marrow mesenchymal stem cells (BMSCs)-尾-tricalcium phosphate tissue engineering bone was implanted at bone defect in blank control group. X-ray, immunohistochemistry and fluorescence quantitative PCR were performed 48 weeks after implantation. Results and conclusion: the X-ray score of sensory nerve group and blood bundle group was higher than that of blank control group (P 0.05), sensory nerve group (P 0.05), sensory nerve group (P < 0.05), and sensory nerve group (P < 0.05). There was no significant difference in X-ray score between vascular bundle group and vascular bundle group.
