柚皮苷對(duì)椎間盤退變的作用及其機(jī)制研究

【摘要】:目的:研究柚皮苷對(duì)體外椎間盤退變模型的作用,以及相應(yīng)的作用機(jī)制。 方法:實(shí)驗(yàn)所需的大鼠髓核細(xì)胞來自于大鼠的尾椎椎間盤。利用IL-1β作用于髓核細(xì)胞建立椎間盤退變的細(xì)胞模型。用不同濃度的柚皮苷作用于處理后的髓核細(xì)胞,對(duì)比它對(duì)基質(zhì)金屬蛋白酶表達(dá)的影響,以及對(duì)于轉(zhuǎn)錄因子NF-κB及其通路的影響。 結(jié)果:柚皮苷能夠明顯降低由IL-1β刺激髓核細(xì)胞的基質(zhì)金屬蛋白酶的表達(dá),并能夠通過提高IκB-α的表達(dá)水平來抑制NF-κB通路的活化。 結(jié)論:柚皮苷是一種有效的抗炎和抗分解代謝藥物,通過抑制NF-κB信號(hào)通路在髓核細(xì)胞中發(fā)揮作用,可以被研發(fā)為一種治療椎間盤退變的有效藥物。
[Abstract]:Aim: to study the effect of naringin on the model of disc degeneration in vitro and its mechanism. Methods: the rat nucleus pulposus cells were obtained from the rat caudal disc. The cell model of disc degeneration was established by the action of IL-1 尾 on nucleus pulposus cells. The effects of naringin at different concentrations on the expression of matrix metalloproteinase and the transcription factor NF- 魏 B and its pathway were compared. Results: naringin could significantly decrease the expression of matrix metalloproteinases stimulated by IL-1 尾 and inhibit the activation of NF- 魏 B pathway by increasing the expression of I 魏 B- 偽. Conclusion: naringin is an effective anti-inflammatory and anti-catabolism drug. It can be developed as an effective drug for the treatment of intervertebral disc degeneration by inhibiting NF- 魏 B signaling pathway in nucleus pulposus cells.
