六磷酸肌醇對(duì)人骨肉瘤細(xì)胞株MG-63和原代培養(yǎng)成骨細(xì)胞增殖的影響

【摘要】:目的研究六磷酸肌醇(inositol hexaphosphate,IP6)對(duì)人骨肉瘤細(xì)胞株MG-63和原代培養(yǎng)成骨細(xì)胞增殖的影響,并探求其抗癌效果的最佳作用濃度。方法原代培養(yǎng)人成骨細(xì)胞并鑒定,復(fù)蘇并常規(guī)培養(yǎng)人骨肉瘤細(xì)胞株MG-63,MTT法檢測(cè)不同濃度IP6干預(yù)培養(yǎng)時(shí)兩種細(xì)胞的增殖情況,確定IP6的最佳抑癌濃度,并進(jìn)一步用流式細(xì)胞儀檢測(cè)MG-63細(xì)胞周期及凋亡情況。結(jié)果當(dāng)IP6濃度≥1mmol/L時(shí)開始抑制MG-63細(xì)胞增殖,在4mmol/L時(shí)達(dá)到最大,并呈時(shí)間-劑量依賴性;當(dāng)IP6濃度為0.5、1mmol/L時(shí),對(duì)成骨細(xì)胞增殖抑制不明顯,2mmol/L時(shí)輕度抑制,4mmol/L時(shí)導(dǎo)致成骨細(xì)胞凋亡。結(jié)論 IP6能夠抑制人骨肉瘤細(xì)胞MG-63的增殖并導(dǎo)致其凋亡,其抗癌的最佳作用濃度為2mmol/L。
