七氟烷復(fù)合咪達(dá)唑侖麻醉對老年大鼠認(rèn)知功能的影響及其機制

【摘要】:目的探討七氟烷復(fù)合咪達(dá)唑侖麻醉對老年大鼠認(rèn)知功能的影響及其機制。方法將64只SD大鼠隨機分為對照組(C組)、七氟烷組(S組)、咪達(dá)唑侖組(M組)、七氟烷復(fù)合咪達(dá)唑侖組(S+M組),每組大鼠16只,處理后采用Morris水迷宮實驗檢測各組大鼠認(rèn)知功能的變化以及各組大鼠海馬N-甲基-D-天冬氨酸受體(NR)1、NR2B mRNA表達(dá)情況、各組大鼠海馬神經(jīng)型一氧化氮合酶(n NOS)、半胱氨酸蛋白酶(caspase)-3蛋白表達(dá)。結(jié)果 Morris水迷宮實驗訓(xùn)練1 d,各組大鼠潛伏期比較差異無統(tǒng)計學(xué)意義(P0.05),訓(xùn)練3 d、5 d,各麻醉組大鼠潛伏期均顯著高于C組,差異有統(tǒng)計學(xué)意義(P0.05),且S+M組大鼠潛伏期明顯高于S組及M組(P0.05),差異有統(tǒng)計學(xué)意義;將平臺撤除后,各麻醉組大鼠60 s內(nèi)穿過平臺次數(shù)明顯低于C組,差異有統(tǒng)計學(xué)意義(P0.05),且S+M組大鼠穿過平臺次數(shù)明顯低于S組及M組,差異有統(tǒng)計學(xué)意義(P0.05);各組大鼠海馬NR1 mRNA相對表達(dá)量比較差異無統(tǒng)計學(xué)意義(P0.05),S組與S+M組大鼠海馬NR2B mRNA相對表達(dá)量較C組、M組顯著降低,差異有統(tǒng)計學(xué)意義(P0.05),M組與C組大鼠海馬NR2B mRNA相對表達(dá)量差異無統(tǒng)計學(xué)意義(P0.05);與C組比較,各麻醉組大鼠海馬n NOS蛋白表達(dá)降低,差異有統(tǒng)計學(xué)意義,且S+M組大鼠海馬n NOS蛋白表達(dá)量最低,而各組大鼠海馬caspase-3蛋白表達(dá)情況無明顯變化,差異有統(tǒng)計學(xué)意義(P0.05)。結(jié)論七氟烷復(fù)合咪達(dá)唑侖可引起大鼠認(rèn)知功能的降低,其機制可能與抑制NR2B、NOS表達(dá)有關(guān),而與神經(jīng)細(xì)胞凋亡無明顯關(guān)系。
[Abstract]:Objective to investigate the effect and mechanism of sevoflurane combined with midazolam anesthesia on cognitive function in aged rats.Methods Sixty-four SD rats were randomly divided into control group (C), sevoflurane group (group S), midazolam group (group M), sevoflurane combined with midazolam group (group S M), and sevoflurane combined with midazolam group (n = 16).After treatment, the changes of cognitive function and the expression of N- methyl-Daspartic acid receptor (NR1) NR2B mRNA in hippocampus of rats in each group were detected by Morris water maze test.The expression of neuronal nitric oxide synthase (nNOS) and cysteine protease (Caspase-3) in hippocampus of rats in each group.Results Morris water maze experiment training for 1 day, there was no significant difference in the latency of each group (P 0.05), training 3 days and 5 days, the latency of each anesthetic group was significantly higher than that of C group.The latency of rats in S M group was significantly higher than that in S group and M group (P 0.05).The difference was statistically significant (P 0.05), and the times of crossing the platform in S M group were significantly lower than those in S group and M group.There was no significant difference in the relative expression of NR1 mRNA between the two groups. The relative expression of NR2B mRNA in the hippocampus of S group and S M group was significantly lower than that of C group.There was no significant difference in the relative expression of NR2B mRNA between group M and group C (P 0.05), but the expression of n NOS protein in hippocampus of all anesthetized groups was lower than that of group C (P 0.05).The expression of n NOS protein in hippocampus of S M group was the lowest, but the expression of caspase-3 protein in hippocampus of all groups had no significant change (P 0.05).Conclusion sevoflurane combined with midazolam can induce the decrease of cognitive function in rats, and its mechanism may be related to the inhibition of NOS expression in NR2BNs, but not to neuronal apoptosis.
