正常與退變髓核細(xì)胞對(duì)骨髓間充質(zhì)干細(xì)胞誘導(dǎo)分化的對(duì)比研究

【摘要】:目的正常髓核細(xì)胞(nNPCs)對(duì)骨髓間充質(zhì)干細(xì)胞(MSCs)有誘導(dǎo)分化作用,而退變髓核細(xì)胞(dNPCs)對(duì)MSCs的誘導(dǎo)作用是否存在差異尚未闡明,文中對(duì)比nNPCs與dNPCs對(duì)MSCs向髓核細(xì)胞分化誘導(dǎo)作用的差異。方法細(xì)胞株/退變患者來源的人NPCs與細(xì)胞株來源人MSCs行體外三維共培養(yǎng),分為5組:nNPCs對(duì)照組、dNPCs對(duì)照組、MSCs對(duì)照組、nNPCs-MSCs組(nNPCs誘導(dǎo)的MSCs)、dNPCs-MSCs組(dNPCs誘導(dǎo)的MSCs),共培養(yǎng)7 d后,行RT-PCR及Western blot檢測(cè)各組細(xì)胞聚集蛋白聚糖(ACAN)、Ⅱ型膠原(COL-2)、SOX-9、基質(zhì)金屬蛋白酶-1(MMP-1)、MMP-3的表達(dá)水平。結(jié)果 RTPCR結(jié)果顯示,與nNPCs對(duì)照組相比:dNPCs對(duì)照組ACAN、COL-2、SOX-9的表達(dá)較低(P0.05),MMP-1、MMP-3的表達(dá)相對(duì)較高(P0.01);nNPCs-MSCs組ACAN的表達(dá)相對(duì)較低(P0.05);dNPCs-MSCs組ACAN、COL-2、SOX-9的表達(dá)相對(duì)較低(P0.05)。與dNPCs對(duì)照組相比:nNPCs-MSCs組ACAN、COL-2、SOX-9的表達(dá)相對(duì)較高(P0.05),MMP-1、MMP-3的表達(dá)較低(P0.01);dNPCs-MSCs組MMP-1、MMP-3的表達(dá)較低(P0.01)。與MSCs對(duì)照組相比:nNPCs-MSCs組及dNPCs-MSCs組ACAN、COL-2、SOX-9的表達(dá)均相對(duì)較高(P0.01)。與nNPCs-MSCs組相比:dNPCs-MSCs組ACAN、COL-2、SOX-9的表達(dá)相對(duì)較低(P0.05)。Western blot結(jié)果顯示:與nNPCs對(duì)照組相比:dNPCs對(duì)照組ACAN、COL-2、SOX-9的表達(dá)較低(P0.05),而MMP-1、MMP-3的表達(dá)較高(P0.05);nNPCs-MSCs組MMP-1、MMP-3的表達(dá)相對(duì)較低(P0.01);dNPCs-MSCs組ACAN、COL-2、SOX-9、MMP-1、MMP-3的表達(dá)較低(P0.05)。與dNPCs對(duì)照組相比:nNPCs-MSCs組ACAN、COL-2、SOX-9的表達(dá)較高(P0.05),MMP-1、MMP-3的表達(dá)較低(P0.01);dNPCs-MSCs組MMP-1、MMP-3的表達(dá)較低(P0.01)。與MSCs對(duì)照組相比:nNPCs-MSCs組及dNPCs-MSCs組ACAN、COL-2、SOX-9的表達(dá)均相對(duì)較高(P0.01)。與nNPCs-MSCs組相比:dNPCs-MSCs組ACAN、COL-2、SOX-9的表達(dá)均相對(duì)較低(P0.05)。結(jié)論 nNPCs及dNPCs均能誘導(dǎo)MSCs向髓核樣細(xì)胞分化,與nNPCs共培養(yǎng)后,MSCs的ACAN、COL-2、SOX-9的表達(dá)水平與nNPCs相當(dāng),優(yōu)于同樣培養(yǎng)條件下dNPCs對(duì)MSCs的誘導(dǎo)。
[Abstract]:The normal nucleus pulposus cells (nNPCs) on bone marrow mesenchymal stem cells (MSCs) induced differentiation and degeneration of the nucleus pulposus cells (dNPCs) induced by MSCs has not yet been clarified whether there are differences between nNPCs and dNPCs, this paper induces the differentiation of MSCs into nucleus pulposus cells.
