川芎嗪對神經(jīng)病理性疼痛大鼠背根神經(jīng)節(jié)GAP-43mRNA表達(dá)的影響

【摘要】:目的:評價川芎嗪對慢性坐骨神經(jīng)壓迫性損傷大鼠的脊髓背根神經(jīng)節(jié)GAP-43 mRNA表達(dá)的影響,初步探討川芎嗪對神經(jīng)病理性疼痛大鼠的作用和相關(guān)的機(jī)制。方法:選取成年雄性健康Wistar大鼠共54只,周齡6~8周,體重180~220 g。分組方法采用隨機(jī)數(shù)字表法,即將54只大鼠分為3組(n=18):假手術(shù)組(S組)、神經(jīng)病理性疼痛組(NP組)和川芎嗪組(TMP組)。NP組和TMP組采用慢性坐骨神經(jīng)壓迫損傷法建立神經(jīng)病理性疼痛模型,S組大鼠僅暴露坐骨神經(jīng)不結(jié)扎。TMP組于CCI術(shù)畢即刻腹腔注射川芎嗪100mg·kg-1,1天1次,而S組和NP組給予等容量生理鹽水腹腔注射,連續(xù)給藥至處死當(dāng)天。S組、NP組和TMP組分別隨機(jī)取6只大鼠,于術(shù)前1 d,術(shù)后3、7、14 d(T0~3)時測定機(jī)械痛閾(MWT)和熱痛閡(TWL),并于T1-3時測定痛閾后處死,取術(shù)側(cè)L4-L6背根神經(jīng)節(jié)(DRG),采用實(shí)時熒光PCR法檢測生長相關(guān)蛋白(GAP-43)mRNA的表達(dá)水平。結(jié)果:與S組比較,NP組和TMP組MWT和TWL在T1-3時降低,背根神經(jīng)節(jié)GAP-43 mRNA表達(dá)上調(diào)(P0.05)。與NP組比較,TMP組MWT和TWL在T1-3時升高,背根神經(jīng)節(jié)GAP-43 mRNA表達(dá)下調(diào)(P0.05)。與T1時比較,NP組和TMP組MWT和TWL在T2-3時升高,背根神經(jīng)節(jié)GAP-43mRNA表達(dá)下調(diào)(P0.05)。結(jié)論:川芎嗪減輕大鼠神經(jīng)病理性疼痛的機(jī)制可能與其抑制脊髓背根神經(jīng)節(jié)GAP-43 mRNA表達(dá)上調(diào)有關(guān)。
[Abstract]:Aim: to evaluate the effect of ligustrazine on the expression of GAP-43 mRNA in spinal dorsal root ganglion (DRG) of rats with chronic sciatic nerve compression injury, and to explore the effect of ligustrazine on neuropathic pain in rats.Methods: 54 adult male healthy Wistar rats were selected.The grouping method is random digital table,54 rats were divided into 3 groups: sham operation group (S group), neuropathic pain group (NP group), tetramethylpyrazine group (TMP group) and TMP group (using chronic sciatic nerve compression injury) to establish neuropathic pain model rats in S group.In the unligated sciatic nerve group, ligustrazine 100mg kg-1 was injected intraperitoneally immediately after CCI.Group S and group NP were intraperitoneally injected with normal saline of equal volume, and 6 rats were randomly selected from group S and group TMP, respectively, on the day of death.The mechanical pain threshold (MWTT) and thermal pain barrier (TWL) were measured 1 day before operation and 7 days after operation, and the pain threshold was killed at T1-3. The expression of growth associated protein (GAP-43) was detected by real-time fluorescence PCR method in the dorsal root ganglion (DRG) of the dorsal root ganglion of L4-L6.Results: compared with S group, MWT and TWL in NP group and TMP group were decreased at T1-3, and GAP-43 mRNA expression was up-regulated in dorsal root ganglion (DRG).Compared with NP group, MWT and TWL in TMP group increased at T1-3 and the expression of GAP-43 mRNA in DRG was down-regulated (P 0.05).Compared with T1, MWT and TWL in NP group and TMP group were increased at T2-3, and the expression of GAP-43mRNA in dorsal root ganglion was down-regulated (P 0.05).Conclusion: the mechanism of ligustrazine in relieving neuropathic pain may be related to the up-regulation of GAP-43 mRNA expression in dorsal root ganglion of spinal cord.
