槲皮素對(duì)兔骨關(guān)節(jié)炎模型關(guān)節(jié)液中IL-1β、TNFα水平的影響

【摘要】:目的:觀察目前槲皮素對(duì)骨關(guān)節(jié)炎動(dòng)物模型關(guān)節(jié)液中細(xì)胞因子IL-1β、TNF-α水平的影響。方法:將20只新西蘭大白兔按照hulth法建立兔膝關(guān)節(jié)骨關(guān)節(jié)炎模型,隨機(jī)等分成4組,每組各5只。(1)槲皮素組(2)塞來(lái)昔布組(3)生理鹽水組(4)模型組。建模4周后各治療組分別給予藥物灌胃治療,槲皮素組建模開(kāi)始灌槲皮素10.31mg/(kg·d),塞來(lái)昔布組10.31mg/(kg·d),生理鹽水組10.31ml/(kg·d),模型組不給予藥物灌胃治療,連續(xù)灌胃4周。4周后給予空氣栓塞處死,抽取膝關(guān)節(jié)液用ELISA法檢測(cè)關(guān)節(jié)液中IL-1β、TNF-α的含量,并取股骨內(nèi)髁關(guān)節(jié)軟骨標(biāo)本送普通病理檢查及做膝關(guān)節(jié)大體評(píng)分。結(jié)果:(1)模型組股骨內(nèi)髁處可見(jiàn)軟骨表面軟骨剝脫,深達(dá)骨質(zhì),關(guān)節(jié)內(nèi)有骨贅形成。HE染色可見(jiàn)骨組織及軟骨細(xì)胞較少。(2)槲皮素、塞來(lái)昔布治療組膝關(guān)節(jié)大體評(píng)分顯著低于模型組及生理鹽水組(P0.05),槲皮素、塞來(lái)昔布組間大體評(píng)分無(wú)差異(P0.05)。(3)槲皮素組、塞來(lái)昔布組膝關(guān)節(jié)液中IL-1β、TNF-α水平顯著低于模型組及生理鹽水組(P0.05)。結(jié)論:槲皮素、塞來(lái)昔布均能夠抑制OA模型關(guān)節(jié)軟骨破壞及關(guān)節(jié)滑液中IL-1β、TNF-α的水平。
[Abstract]:Objective: to observe the effect of quercetin on the level of cytokine IL-1 尾 TNF- 偽 in the articular fluid of osteoarthritis animal model. Methods: twenty New Zealand white rabbits were randomly divided into 4 groups according to the hulth method to establish rabbit knee osteoarthritis model. Each group (n = 5) quercetin group (n = 2) celecoxib group (n = 3) normal saline group (n = 4). Quercetin group was treated with quercetin 10.31mg/(kg dronium, celecoxib group with 10.31mg/(kg dronium and normal saline group with 10.31ml/(kg dronium. The model group was given no drug infusion and was killed by air embolization for 4 weeks. The content of IL-1 尾 -TNF- 偽 in articular fluid was determined by ELISA method in knee joint fluid. The specimens of femoral medial condylar articular cartilage were taken for general pathological examination and knee joint gross score. Results cartilage surface cartilage peeling was seen in the femoral medial condyle in the model group. Osteophyte formation. He staining showed that there were fewer bone tissues and chondrocytes. Quercetin. The gross score of knee joint in celecoxib treated group was significantly lower than that in model group and saline group (P 0.05), quercetin, and quercetin. There was no difference in gross score between celecoxib group (P 0.05) Quercetin group and celecoxib group had significantly lower IL-1 尾 -TNF- 偽 level in knee joint fluid than model group and normal saline group (P 0.05). Conclusion: quercetin, celecoxib group, celecoxib group, celecoxib group, celecoxib group and normal saline group, Celecoxib could inhibit the destruction of articular cartilage and the level of IL-1 尾 -TNF- 偽 in synovial fluid of OA model.
