丙泊酚后處理對大鼠肝缺血再灌注時腎組織細胞凋亡Bcl-2和Bax的影響

【摘要】:目的:研究丙泊酚后處理對大鼠肝臟缺血再灌注時腎臟細胞B淋巴細胞瘤/白血病-2(Bcl-2),Bcl相關X蛋白(Bax)的影響。方法:健康雄性SD大鼠54只,隨機分成3組:假手術(S)組,缺血再灌注(IR)組,丙泊酚(P)組,(n=18)每組再根據(jù)再灌注時間(2h、4h、6h)的不同分成3個時相,每個時相為一個亞組,每個亞組各6只大鼠。分別為S2、S4、S6、IR2、IR4、IR6、P2、P4、P6亞組,共九個亞組。完成制作約70%肝缺血再灌注的模型,除S組之外,其余各組均為缺血45min后恢復灌注,S組、IR組缺血35min時以0.3ml·kg-1·min-1持續(xù)靜脈泵注0.9%生理鹽水,P組缺血35min時以0.3ml·kg-1·min-1持續(xù)泵注0.1%丙泊酚溶液。泵注時間均為10min,即3mg/kg。實驗結(jié)束即刻取大鼠左腎下極,行組織切片和蘇木精-伊紅(HE)染色,光鏡下觀察。檢測各樣本大鼠血清中半胱氨酸蛋白酶抑制劑C(Cys-c)的濃度。使用免疫組化方法檢測各組腎組織Bcl-2和Bax蛋白含量的表達,TUNEL法計算腎臟細胞的凋亡指數(shù)(AI)。結(jié)果:1、與S組相比,IR組和P組的Bcl-2、Bax表達增多(P0.05);與IR組相比,P組Bcl-2蛋白表達增多,Bax蛋白表達減少(P0.05)。2、與S組相比,IR組和P組的AI值均增高(P0.05);與IR組相比,P組的AI值減少(P0.05)。3、與S組相比,IR組和P組的Cys-c含量均增高(P0.05);與IR組相比,P組的Cys-c含量減少(P0.05)。4、光鏡下IR組腎組織病理損傷變化比P組明顯。結(jié)論:1、丙泊酚對肝缺血再灌注后不同時刻的腎臟損傷具有保護作用。2、這種保護機制可能與調(diào)控Bcl-2,Bax蛋白的表達有關。
[Abstract]:Objective: to study the effect of propofol postconditioning on renal B lymphocyte tumor / leukaemia Bcl-2Bcl related X protein Bax.Methods: 54 healthy male SD rats were randomly divided into 3 groups: sham operation group. Each group was divided into 3 time phases according to the reperfusion time of 2 h ~ 4 h ~ (6 h). Each phase was a subgroup with 6 rats in each subgroup. There were nine subgroups. About 70% liver ischemia-reperfusion model was made, except group S, The rest groups were treated with 0.3ml kg-1 min-1 continuous infusion of 0.1% propofol solution during 35min ischemia with 0.3ml kg-1 min-1 at the end of the experiment. The injection time was 10 min, that is, 3 mg / kg. The left subrenal pole of the rats was taken immediately after the end of the experiment, and the rest groups were injected with 0.3ml kg-1 min-1 continuously at the end of the experiment, and the rats were injected with 0.1% propofol solution at the end of the experiment. Tissue sections and hematoxylin-eosin (HEH) staining were performed. The concentration of cysteine protease inhibitor (Cy Cys-c) in serum of rats was detected under light microscope. The expressions of Bcl-2 and Bax protein in renal tissues were detected by immunohistochemical method. The apoptotic index of renal cells was calculated by Tunel method. Results compared with S group and P group, the expression of Bcl-2nBax protein increased and the expression of Bcl-2 protein decreased in P group compared with S group, and the AI value in P group and P group was higher than that in group S and P group (P 0.05), and the AI value in P group was lower than that in P group (P < 0.05), while that in P group was significantly higher than that in P group (P < 0.05), the expression of Bcl-2 protein in P group was higher than that in group S and P group (P < 0.05). Compared with group S, the content of Cys-c in IR and P groups was higher than that in group S, and the content of Cys-c in group P was lower than that in group IR. The pathological changes of renal tissue in IR group were more obvious than that in group P. Conclusion 1 and propofol have different effects on hepatic ischemia-reperfusion injury. This protective mechanism may be related to the regulation of the expression of Bcl-2P Bax protein.
