正中神經(jīng)電刺激對(duì)腦外傷后昏迷大鼠前額葉皮質(zhì)區(qū)NMDA NR1蛋白表達(dá)變化的影響

【摘要】:目的:研究正中神經(jīng)電刺激對(duì)腦外傷昏迷大鼠前額葉皮質(zhì)區(qū)(prefrontal cortex,PFC)NMDA NR1受體水平的影響。方法:將144只SD大鼠隨機(jī)分為空白組,假刺激組,刺激組和OX1R拮抗劑組共4組。采用經(jīng)典的自由落體模型建立腦外傷昏迷模型及大鼠意識(shí)狀態(tài)6級(jí)評(píng)定標(biāo)準(zhǔn)評(píng)估四組大鼠行為學(xué)變化,并于實(shí)驗(yàn)完成后6,12,24小時(shí)處死大鼠,分別用Western-blot及免疫組織化技術(shù)檢測(cè)各組大鼠前額葉皮質(zhì)組織的NMDA NR1受體表達(dá)。結(jié)果:刺激組25只大鼠(25/36)出現(xiàn)翻正反射,OX1R拮抗劑組17只(17/36)、假刺激組11只(11/36)出現(xiàn)翻正反射。Western-blot法結(jié)果顯示四組NMDA NR1受體水平在6,12,24h三個(gè)時(shí)間點(diǎn)均呈現(xiàn)“OX1R拮抗劑組假刺激組空白組刺激組”遞增趨勢(shì),且差異有統(tǒng)計(jì)學(xué)意義(P0.05),而同一組不同時(shí)間點(diǎn)的比較表現(xiàn)為:空白組及假刺激組:“6h12h24h”,差異有統(tǒng)計(jì)學(xué)意義(P0.05);刺激組:“6h12h24h”,差異無(wú)統(tǒng)計(jì)學(xué)意義(P㧐0.05);OX1R拮抗劑組:“6h24h12h”,差異有統(tǒng)計(jì)學(xué)意義(P0.05)。免疫組化結(jié)果顯示組間比較前額葉組織NMDA NR1受體表達(dá)呈“OX1R拮抗劑組假刺激組空白組刺激組”遞增趨勢(shì)(P0.05),組內(nèi)比較呈“6h12h24h”趨勢(shì)(P㧐0.05)。結(jié)論:正中神經(jīng)電刺激可作為一種治療腦外傷后昏迷的有效促醒方法,其機(jī)制可能與前額葉皮質(zhì)NMDA NR1受體水平上調(diào)有關(guān),且Orexin-A可能在其中發(fā)揮調(diào)節(jié)作用。
[Abstract]:Objective: to study the effect of median nerve electrical stimulation on the level of NMDA NR1 receptor in prefrontal cortex of prefrontal cortex of rats with brain injury. Methods: 144 SD rats were randomly divided into blank group and sham stimulation group. The rats in the stimulation group and the OX1R antagonist group were divided into 4 groups. The model of brain trauma coma was established by the classical free-fall body model and the rats' consciousness status was assessed by the 6-grade assessment standard. The rats were killed at 61212h 24 hours after the experiment, and the rats were killed 24 hours after the experiment. The expression of NMDA NR1 receptor in prefrontal cortex of rats in each group was detected by Western-blot and immunohistochemistry respectively. Results: in the stimulation group, 25 / 36 rats had positive and positive reflex OX1R antagonist group (17 / 17 / 36) and the sham stimulation group (11 / 11 / 36) had positive and negative inversion. Western-blot analysis showed that the level of NMDA NR1 receptor in the four groups showed an increasing trend at the three time points of 6 ~ 1212 ~ 24 h, "OX1R antagonist group, sham stimulation group, blank stimulation group", and "OX1R antagonist group, sham stimulation group, blank stimulation group". The difference was statistically significant (P 0.05), but the difference between blank group and sham stimulation group was "6 h 12 h 24 h" (P 0.05), while that of stimulation group was "6 h 12 h 24 h" (P < 0.05), but there was no significant difference between control group and sham stimulation group (P < 0.05). The results of immunohistochemistry showed that the expression of NMDA NR1 receptor in prefrontal lobe tissue of OX1R antagonist group showed an increasing trend of increasing (P0.05N), while the expression of NR1 receptor in prefrontal lobe tissue showed a trend of "6h 12h 24 h" in the control group of OX1R antagonist group and "6h 12h 24 h" in the group with P0. 05% OX1R antagonist, and there was a tendency of P0. 05% expression in the prefrontal lobe tissue of the group compared with that of the control group (P 0. 05%). Conclusion: median nerve stimulation can be used as an effective method for the treatment of coma after brain injury. The mechanism may be related to the up-regulation of NMDA NR1 receptor in prefrontal cortex, and Orexin-A may play a regulatory role in it.
