【摘要】：骨肉瘤是好發(fā)于兒童及青少年的惡性骨腫瘤,由于骨組織硬度大、滲透性差和生理生化過程的特殊性,抗腫瘤藥物在骨組織很難達(dá)到有效的藥物濃度。選擇增加藥物劑量不僅提高了藥物毒副反應(yīng)的發(fā)生率,也給患者在治療過程中帶來心理及經(jīng)濟(jì)上的壓力。實(shí)驗(yàn)研究發(fā)現(xiàn)了多種途徑能使抗腫瘤藥物通過自我導(dǎo)向或在外界環(huán)境誘導(dǎo)下選擇性地與腫瘤組織結(jié)合,其中包括通過骨誘導(dǎo)劑(雙膦酸鹽類)偶聯(lián)化療藥物、納米藥物載體封裝化療藥物以及小分子核糖核酸結(jié)合化療藥物等方式。本文就目前國內(nèi)外研究抗腫瘤藥物通過靶向給藥系統(tǒng)治療骨肉瘤的實(shí)驗(yàn)成果、理論機(jī)制及治療方向作綜述。
[Abstract]:Osteosarcoma is a malignant bone tumor that occurs in children and adolescents. Due to its high bone hardness, poor permeability and particularity of physiological and biochemical processes, osteosarcoma is a malignant tumor in children and adolescents. Antitumor drugs are difficult to achieve effective drug concentration in bone tissue. Choosing to increase the dosage of drugs not only increases the incidence of adverse drug reactions, It also brings psychological and economic pressure to patients during the course of treatment. Experimental studies have found that many ways can enable anti-tumor drugs to selectively bind to tumor tissue through self-directed or induced by external environment. These include coupling chemotherapy drugs with bone inducers (bisphosphonates), Nano-drug carriers encapsulate chemotherapeutic drugs and small molecule ribonucleic acid combined with chemotherapeutic drugs. The theoretical mechanism and therapeutic direction are reviewed.
